摘 要:目的研究大肠癌中肿瘤特异性抗原MAGE-A1,Ki-67的表达及其相关性,探讨他们与大肠癌生物学行为的关系及其作为免疫治疗靶点的可行性。方法免疫组化S-P法,利用鼠抗人MAGE-A1、Ki-67单克隆抗体检测60例大肠癌标本中MAGE-A1,Ki-67表达水平。结果60例大肠癌中MAGE-A1表达率为33.3%(20/60),高表达率为5%(3/60),低表达率为28.3%(17/60),正常组织表达率为3.33%,大肠癌和正常组织之间的表达具有显著性差异(P(0.05)。Ki-67表达率为53.3%(32/60),高表达率为33.3%(20/60),低表达率为20%(12/60),正常组织表达率为16.6%(4/30),大肠癌和正常组织之间的表达具有显著性差异(P(0.05)。MAGE-A1与分化程度、淋巴结转移有关(P(0.05),Ki-67与分化程度有关(P(0.05)。MAGE-A1阳性表达的高低与Ki-67阳性表达的高低相关(r=0.384,P(0.05)。结论MAGE-A1在大肠癌中有较高的表达,并且与细胞增殖相关,正常组织不表达,MAGE-A1阳性患者的预后好,有望作为免疫治疗的靶点。Objective This experiment aims at investigating the expression of MAGE-A1 and Ki-67 in colorectal cancer and their relationship , discussing the relationship with the clinical pathological features ,investigating the initial possibility for immune therapy. Methods 60 patients of colorectal cancer were examined for MAGE-A1 , Ki-67 by using SP immunohistochemical technique on formalin-fixed , paraffin-embedded sections . Results The positive rate of MAGE-A1 was 33.3% (20/60), high expression rate was 5% (3/60), and low expression rate was 28.3% (44/60), the positive rate in colorectal cancer was significantly higher than that in normal tissues (3.33%). The positive rate of Ki-67 was 53.3% (56/60) with high expression rate of 33.3% (20/60), and low expression rate of 20% (12/60). The positive rate in colorectal cancer was significantly high- er than that in normal tissues 5% (5/30). There was a relationship with the positive expression of MAGE-A1 and Ki-67 ( r = 0.384, P 〈 0.05 ). MAGE-A1 was correlated to differentiated degree and lymph node metastasis ( P 〈 0.05), Ki-67 was correlated to differentiated degree (P 〈0.05). Conclusion MAGE-A1 expression was cor- related with proliferation of tumor ceils in colorectal cancer. The prognosis of patients with positive expression of MAGE-A1 is good. MAGE-AL may serve as a potential target for immune therapy.
