摘 要:目的研究单涎酸神经节苷脂对大鼠大脑中动脉缺血再灌注损伤的保护作用。方法64只SD大鼠随机分成A组(正常组,11只)、B组(假手术组,11只)、C组(NS对照组,14只)、D组(GM1治疗组,14只)、E组(GM1预防组,14只)。采用改良"线栓法"制作大鼠MCAO再灌注模型,不同方案尾静脉注射GM1。采用"盲法"分别于再灌注10min和再灌注24h对大鼠进行神经功能缺失评分;处死大鼠,A、B组3只,C、D、E组6只大鼠进行TTC染色测量脑梗死体积;各组6只大鼠用作HE染色光镜观察;各组2只大鼠用作电镜观察。结果MCAO再灌注10min评分结果,C、D、E组>A、B组(P<0.01),C、D组>E组(P<0.01);MCAO再灌注24h评分结果,C组>A、B、E组(P<0.01),D组>A、B、E组(P<0.05),C组>D组(P<0.05);脑梗死体积比C、D、E组>A、B组(P<0.01),C组>D、E组(P<0.01),D组>E组(P<0.05);HE染色见梗死灶中心区神经细胞坏死,周围区细胞不同程度损伤;电镜观察A、B组细胞完整,超微结构清晰,C组细胞超微结构破坏严重,D、E组细胞超微结构破坏程度轻。结论GM1能减小MCAO再灌注后脑梗死体积,改善神经功能障碍;GM1对大鼠MCAO再灌注损伤有保护作用。Objective To explore the neuroprotecting effect of monosialo- ganglioside (GM1) on the middle cerebral artery occlusion reperfusion in rats. Methods The SD rats were marked to group A B C D E by randomly. Group A was the normal; group B was sham-operation; group C was injected by normal saline (NS) after MCAO reperfusion; group D was injected by GM1 after MCAO and group E was injected by GM1 24 hours in advance be fore the operation and injected after MCAO. The rats were killed at 24h after MCAO reperfusion. Brains were re moved, respectively, to be stained with TIC and calculated theinfarct volume ratios. Every rat in the study was evaluated the neurobehavioral function at 10min and 24h after MCAO repeffusion. Results The neurobehavioral function at 10min of group E was the lowest among group C D E ( P 〈 0.05), but it was higher than those in group A and B ( P 〈 0.01). The neurobehavioral function at 24h of group E was the lowest among group C D E ( P 〈 0.05), there was no difference among group A B E ( P 〉 0.05), Group D was lower than group C ( P 〈0. 05). The infarct volume ratios of group C D E were higher than those in group A and B ( P 〈 0.05). The infarct volume ratios of group D E were lower than those in group C ( P 〈 0.05) ; The infarct volume ratios of group E was lower than those in group D ( P 〈 0.05). Observing the micro structure of the neurons by TEM, the results showed tha the micro structure of the neurons were normal in group A&B, injured heavily in group C and injured slightly in group D&E. Conclusion The application of GM1 can provide protection for the injury induced by MCAO reperfusion. It can reduce infarct volume and decrease neurological impairment.
