摘 要:目的:观察舌鳞癌Tca-8113细胞系的单细胞体外增殖能力,寻找肿瘤干细胞存在的依据,从而为鉴定其表面标记物奠定理论基础。方法:采用有限稀释法对舌鳞癌Tca-8113细胞进行单细胞体外培养,了解舌鳞癌细胞(Tca-8113)的分裂、增殖特点。结果:单细胞培养8d后,舌鳞癌Tca-8113细胞株中具有持续增殖能力的细胞占肿瘤细胞的10.85%,12d后,继续分裂增殖的细胞比例为5.23%,16d后,仅有5.09%的细胞继续增殖。结论:舌鳞癌Tca-8113细胞具有异质性,仅有少量细胞具有持续增殖能力,而这部分细胞是否就是舌鳞癌Tca-8113细胞的肿瘤干细胞。还有待进一步的实验研究证实。Objective: To examine the proliferation ability of human tongue squamous carcinoma Tca-8113 cell line in order to find the tumor stem cells and to establish the rationale for identification of tumor stem cells surface markers. Methods: in vitro individual cell culture was employed to observe the proliferating character of Tca-8113 ceils. Results: After eight days, in vitro cultured individual cells revealed that about 10.85% of Tea-8113 cell line underwent mitosis, proliferation and cell cloning; after 12 days, whereas 5.23% of cultured Tca-8113 cells continued go on; when after sixteen days, there was also 5.09% of cells still went on. Conclusions: Tongue squamous carcinoma Tca8113 cells have the characteristics of tumoroheterogenesis, and only a minority of tumor cells which may be tumor stem cells exhibited the capability of sustained growth. The future studies should address the tumor stem cells surface markers.
