摘 要:目的探讨枳椇子、大黄水提液(枳黄方)对急性乙醇性肝损伤大鼠(ALD)的保护作用。方法将30只大鼠随机分为A、B、C三组每组10只,其中A组予枳黄方6.0 ml/kg灌胃,B、C组予6.0 ml/kg生理盐水灌胃;间隔45 min后A、B组予56°白酒14 ml/kg灌胃,C组予50%葡萄糖14 ml/kg灌胃,均为1次/d,连续10 d。最后1次灌胃后禁食14 h,以2%戊巴比妥钠40 mg/kg腹腔注射麻醉大鼠,采集肝组织。羟胺法检测肝脏超氧化物歧化酶(SOD)活力,TBA法检测肝脏丙二醛(MDA)水平;采用RT-PCR技术检测肝脏NADPH氧化酶gp-91phox mRNA及p22phox mRNA表达。结果与B组比较,A组SOD活性显著升高,MDA水平及gp-91phox、p22phox mRNA表达显著降低。结论枳黄方能对抗ALD大鼠肝脏脂质过氧化,可能机制为降低肝脏NADPH氧化酶gp-91phox mRNA及p22phox mRNA表达。Objective To observe the effect of Zhihuang Fang on the levels of lipid peroxidation of liver in rats with acute alcoholic liver injury and its mechanism. Methods 30 rats were divided into A,B,C groups with 10 in each,group A were peformed intragastic administration with Zhihuang Fary decoction, the other two groups were performed with sodium. Then group A and B were treated with alcoholic and group C were treated with 5% GS,the pathological changes in the liver were observed under light microscope with hematoxylin and eosin (HE) staining 10 d later, the level of superoxide dismutase (SOD) and malondialdehyde (MDA) were detected by hydroxylamine and TBA method respectively, gp-91phox mRNA and p22phox mRNA was determined with reverse transcription polymerase chain reaction. Results Compared with the group B, the lever of SOD in the group A increased,level of MDA and expression of gp-91phox,p22phox mRNA decreased significantly. Conclusion Zhihuang Fang can antagonize the lipid peroxidation of the liver in rats with alcoholic liver injury the possible mechanism may decrease the expression of gp-91phox mRNA and p22phox mRNA in liver.
