摘 要:[目的]探讨大气粗颗粒物(PM10)、细颗粒物(PM2.5)对人肺成纤维细胞(HLF)增殖作用的影响和DNA链的损伤作用以及番茄红素的保护作用。[方法]采用细胞增殖试验(MTT)测定PM10、PM2.5以及番茄红素对HLF增殖作用,单细胞凝胶电泳法(SCGE)检测PM10、PM2.5对HLFDNA的损伤程度以及番茄红素对DNA损伤的保护作用。[结果]PM10、PM2.5均能抑制HLF的增殖,当颗粒物质量浓度为50μg/mL时,HLF的细胞存活率下降约10%,随着染毒剂量的增大细胞存活率进一步下降,具有明显的剂量-效应关系(P<0.01)。同样质量浓度下比较,PM2.5对细胞增殖的抑制作用大于PM10;PM10和PM2.5均具有明显的DNA损伤作用,当PMs质量浓度仅为50μg/mL时,即能够引起细胞DNA损伤(P<0.01),且具有明显的剂量-反应关系(P<0.01),但PM2.5的损伤程度小于PM10;番茄红素可以有效地阻止由PM10、PM2.5导致的DNA损伤,浓度为25μmol/L的番茄红素即可明显减轻PMs对HLF造成的DNA损伤。[结论]PMs能抑制HLF的增殖、诱导HLF的DNA链断裂,番茄红素可以有效地减小PMs对HLFDNA造成的损伤。[ Objective ] To investigate the effect on cell proliferation and DNA damage of human lung fibroblasts ( HLF )exposed to ambient coarse ( PM10)and fine (PM2.5 )particle matters and to study the protective effect of lycopene on DNA damage exposed to PM10 and PM2.5. [ Methods] The effects of PM10, PM2.5 and lycopene on HLF proliferation were measured by MTT Assay. The degree of DNA damage exposed to PM10, PM2.5 and the protective effects of lycopene on DNA injury by PM10 and PM2.5 were determined by the single cell gel electrophoresis Assay( SCGE ). [Result] Both PM10 and PM2.5 could inhibit the proliferation of HLF. Cell livability were decreased nearly 10% at 50μg/mL of PM10 and PM2.5, and inhibitive effect of cell proliferation were reduced with significant dose-response relationship ( P 〈 0.01 ). Cell livability in PM2.5 group was lower than that in PM10 group at the same concentration; DNA breaks were obviously increased in HLF exposed to PM10 and PM2.5 above 50 μg/mL, with the significant dose-response relationship ( P 〈 0.01 ). The degree of DNA damage exposed by PM10 was larger than by PM2.5. The effective protection of lycopene against DNA damage induced by PM10 and PM2.5 were observed at 25 μmol/L. [ Conclusion ] PMs could decrease HLF proliferation, and result in DNA chains breakage. Lycopene can efficiently reduce DNA damage induced by PMs.
