摘  要:本文以615近交系小鼠右脚垫皮下注射肝癌细胞Hca-F建立实体瘤动物模型,设空白组、模型组、治疗组和预防组,检测小鼠抑瘤率、脾指数及胸腺指数,进行瘤组织的石蜡切片HE染色及免疫组化SP法检测肿瘤组织Fas和PCNA蛋白表达,研究了硒酸软骨多糖抑制荷瘤小鼠肝癌细胞Hca-F生长的作用。结果表明:硒酸软骨多糖能够明显的抑制肝癌细胞Hca-F的生长,且预防组的抑瘤率显著高于治疗组;通过比较模型组、治疗组和预防组的瘤组织的石蜡切片HE染色,治疗组和预防组的瘤组织大量坏死,且预防组的情况更好于治疗组;治疗组和预防组肿瘤组织的Fas蛋白阳性表达率较模型组Fas阳性表达率明显增高,而治疗组和预防组的PCNA蛋白阳性表达率较模型组明显降低。因此,硒酸软骨多糖一定程度上能够抑制肝癌细胞Hca-F的生长,且在一定程度上预防肝癌的发生,为开发治疗恶性肿瘤的药物提供了选择。To study the inhibitory effect of selenic acid cartilage polysaccharide (CA-Se) on the Hca-F hepatomacarcinoma cell in mice, mice model was established by injecting hepatoma cell line Hca-F ceils into their right axilla. Forty mice were divided into four groups, which were control group, model group, treated group and prevented group. Then the tumor-inhibition rate, spleen index (SI), thymus index (TI) and the HE staining of turnor cells were analyzed and the expression of Fas and PCNA proteins were studied by SP immunity histochemistry method. Result showed that CA-Se can inhibit liver cancer cells Hca-F of the mice, and the tumor-inhibition rate of the prevented group was higher than that of the treated group. The results of HE staining of tumor ceils by paraffin slice also showed that most tumor cells of the treated group and prevented group were necrosis. The Fas protein expression rates of the treated group and prevented group were much higher than that of the model group, while the PCNA protein expression rates of the treated group and prevented group were much lower than that of the model group. It was concluded that CA-Se can inhibit the growth of Hca-F cells in mice, and can be used as a prophylactic agent to hepatocarcinoma.
