摘 要:目的:探讨中心体α、β、γ–微管蛋白在人鳞状上皮宫颈永生化细胞(H8细胞株)和癌细胞(Caski细胞株)的表达差异及其在宫颈癌发生发展过程中的作用。方法:采用免疫细胞化学SP法检测中心体α、β–微管蛋白在人宫颈永生化细胞和癌细胞中的表达差异,提取两株细胞的骨架蛋白并用Western blotting对α、β、γ–微管蛋白的表达水平进行半定量分析。结果:Caski细胞中α、β–微管蛋白表达水平高于H8细胞,差异均有统计学意义(P<0.05)。半定量分析α、β、γ–微管蛋白,Caski细胞中的表达量高于H8细胞。结论:中心体异常可能是宫颈癌细胞恶性表型的特征之一,以中心体微管蛋白为治疗靶点的研究有着可期待的前景。Objective: To investigate the expressions of α-tubulin, β-tubulin and γ-tubulin of centrosome in immortalized hu- man squamous epithelial cell(H8) and cervical cancer cell(Caski) and the role of them in occurrence and development of cervical cancer. Methods: Immunocytochemistry was used to analyze the protein expressions of α-tubnlin and β-tubulin in immortalized human cervical cells and carcinoma cells. The skeleton protein was extracted by ProteoExtract Subcellular Proteome Extraction Kit,and then Western blotting was used to determine the protein expressions of α-tubnlin, β-tubulin and γ-tubulin semi-quantitatively. Results: The protein expressions of α-tubulin, β-tubulin and γ-tubulin in Caski were higher than those in HS(P〈0.05). Conclusion: Centrosome abnormality may be one of the malignant phenotypes of cervical cancer cells. The microtubulin protein as a target in tumor treatment can have a good perspective.
