摘 要:目的(1)观测脑出血大鼠经粒细胞集落刺激因子(GCSF)治疗后的行为学变化,为GCSF治疗脑出血提供行为学支持;(2)验证神经行为学实验Cylinder Test和Narrow-alley Corner Test对神经功能损伤/恢复检测的适用性.方法30只SD大鼠分为三组.GCSF组:10只,先制成脑出血模型,然后给予GCSF治疗;SALINE组:10只,先制成脑出血模型,然后给予生理盐水作为安慰治疗;SHAM组:10只,仅按脑出血模型制作过程施予假手术,不给予其它干预.手术后第1、3、7、11、15天用神经行为学实验Rotarod Test、Cylinder Test和Narrow-alley Corner Test检测三组大鼠的行为学变化,比较结果的异同.结果(1)Rotarod test:手术后第1、3天三组大鼠的运动功能均明显下降,术后第7天开始,GCSF治疗组和SHAM组大鼠运动功能逐渐恢复,第7天时与安慰治疗组(SALINE组)相比差别具有统计学意义(P<0.05),第11天时已恢复至接近手术前的水平.而安慰治疗组运动功能一直处于较低水平;(2)Cylinder Test:SHAM组术后第1天不对称分值(对侧前肢使用率)稍有下降,之后恢复至正常水平;ICH大鼠术后1~7 d不对称分值明显下降(P<0.05),第11天后恢复至正常水平;GCSF组与SALINE组不对称分值无显著性差异;(3)Narrow-alley Corner Test:术后第1天GCSF组大鼠和SALINE组大鼠呈现了高度的不对称分值,之后不对称分值随时间逐渐降低,但恢复缓慢,至第15天时仍未达到正常水平(P<0.05);GCSF组大鼠不对称分值比SALINE组大鼠一直略低,但差别无统计学意义.结论(1)Rotarod Test结果显示GCSF治疗对ICH大鼠早期运动功能康复有促进作用;(2)Cylinder Test能敏感地检测出ICH鼠随时间的神经功能恢复;(3)Narrow-alley Corner Test对神经功能损伤的检测较敏感.Objective (1) To observe the change of behavior of ICH (Intracerebral Hemorrhage) rats which were treated with GCSF (Granuloeyte Colony Stimulating Factor) and to provide ethological support for treating ICH with GCSF. (2) To verify the worthwhile and feasibility of the Cylinder Test and Narrow-alley Corner Test to measure the degree of neural function losing and recovering. Methods 30 SD rats were divided into 3 groups. GCSF group:lO rats were operated into ICH model and then were given GCSF therapy. SALINE group: 10 rats were operated into ICH model and then were given saline therapy. SHAM group:lO rats were only operated according ICH model procedure but were not caused ICH and were not given any therapy. The Rotarod Test, Cylinder Test and Narrow-alley Comer Test were used to measure the behaviors of rats in the 3 groups 1, 3, 7, 11, 15 days after the operation and the results were compared. Results .(1) Rotarod test:the motor function of 3 groups of rats decreased obviously on the 1st and 3rd day after operation. After the 7th day, the motor function of GCSF group and SHAM group recovered gradually. The results of both groups were significantly different (P 〈 0.05) to the one of SALINE group on the 7th day. To the lhh day, the motor function of the GCSF and SHAM group had restored near to the pre-operation level but the one of the SALINE group maintained lower level persistently. (2) Cylinder Test: The asymmetric score the rate of the use of the opposite forelimb of SHAM group slightly decreased on the 1st day and later recovered to the normal level. The asymmetric score of the ICH group decreased significantly (P 〈 0.05) from the 1st to the 7 th day post-operation and restored to normal level after the 1 lth day. The difference of the asymmetric score between the GCSF and SALINE group was unsignificantly. (3) Narrow-alley Comer Test:the GCSF and SALINE group showed high asymmetric score on the 1st day post-operation. Later the score decreased gradually bu
