摘  要:目的观察甲强龙对脊髓源性神经干细胞增殖能力的影响,并探讨其机制。方法体外培养及鉴定C57BL/6小鼠脊髓来源的神经干细胞分为甲强龙组与对照组,通过细胞生长曲线研究甲强龙对其增殖能力的影响,并利用半定量聚合酶链反应(PCR)法及Western blot法观察两组神经干细胞24、48、72h自我更新重要调节因子Bmi-1、Numb、Notch表达的差异。结果细胞生长曲线测定第7天甲强龙组神经干细胞计数为12.4×10^4,而对照组为19.3×10^4,相差6.9×10^4;在24、48、72h甲强龙组与对照组Bmi-1 mRNA表达灰度比值分别为0.52±0.05、0.21±0.02、0.16±0.05,而Numb,Notch mRNA表达无明显变化,Western blot法测定甲强龙组与对照组Bmi-1蛋白表达的灰度比分别为0.63±0.07、0.33±0.04、0.19±0.06。结论甲强龙抑制脊髓源性神经干细胞增殖通过Bmi-1介导。Obiective To study the influence of methylprednisolone (MP) on the proliferation of spinal cord derived neural stem ceils (NSCs) and the action mechanism. Methods NSCs were isolated from the spinal cord of C57BL/6 mice and identified in vitro by immunocytochemistry, and then divided into two groups : MP group and control group. The proliferation of NSCs in these two groups were described by growth curve, and the expression of Bmi-1, Numb and Notch was analyzed by PCR and Western blot at 24, 48 and 72 h. Results At the 7th day of the proliferation study,the number of NSCs in MP and control groups was 12.4 × 10^4 and 19.3 × 10^4 respectively. At 24,48 and 72 h the gray level ratio of Bmi-1 mRNA expression between MP group and control group was 0.52 ±0.05,0.21 ±0.02 and 0.16 ±0.05 respectively. The gray level ratio of the Bmi-1 protein expression between the MP group and control group was 0.63 ± 0.07,0.33 ± 0.04,0.19 ± 0.06 at 24,48 and 72 h, respectively. Conclusion The inhibitory effects of MP on the proliferation of spinal cord derived NSCs was mediated by Bmi-1.
