神经母细胞瘤P-糖蛋白检测及其临床意义

摘  要:①目的检测神经母细胞瘤P-糖蛋白(P-gp)表达情况并探讨其临床意义。②方法用JSB-1P-gp单克隆抗体SLAB法,测定19例神经母细胞瘤石蜡切片的P-gp表达情况。③结果19例神经母细胞瘤有11例P-gp阳性(57.9%)。术前化疗与术前无化疗的神经母细胞瘤P-gp阳性率间差别有显著性(χ2=4.92,P<0.05)。P-gp阳性组神经母细胞瘤病儿的平均生存时间为323d,阴性组为789d,二者具有明显差异(时序检验,P<0.05)。④结论P-gp表达是神经母细胞瘤产生耐药的原因之一。Objective To detect the expression of P glycoprotein in neuroblastoma by immunohistochemical method and to discuss its clinical significance. Methods 19 polyxine coated slides were stained with JSB 1 P glycoprotein monoclonal antibody by SLAB method. Results 11 out of 19 neuroblastomas (57.9%) were positive of P glycoprotein. The positive rate was significantly different between chemotherapeutic and chemotherpy free neuroblastomas preoperatively( χ 2=4.92, P< 0. 05). The average survive time was 323 days in P glycoprotein positive neuroblastoma group and 789 days in the P glycoprotein negative group. The difference between the two groups was significant(log rank test, P< 0.05). Conclusion Detectable P glycoprotein is the cause of drug resistance in neuroblastoma and the positivity of P glycoprotein appears to be an adverse prognostic factor.
