摘 要:目的:探讨乙烷基亚硝基脲(ENU)诱变技术获得的B6-Co小鼠角膜病变的病因。方法:以年龄1~10日的B6-Co小鼠116只为实验组,年龄1日~2个月的88只正常B6小鼠为对照组,取结膜囊和/或角膜病灶刮取物行血平板和营养肉汤增菌培养,并作细菌菌种鉴定。结果:B6小鼠在睁眼前结膜囊细菌培养阴性,睁眼后标本68例,阳性59例(86.76%)。共培养出阳性菌株77株,其中革兰阳性球菌85.71%,革兰阳性杆菌13.00%。缓慢葡萄球菌和里拉微球菌的检出率最高均为36.36%,其次为F-1棒杆菌,说明正常小鼠(睁眼后)结膜囊细菌是外界感染所致;B6-Co小鼠共检标本116只,阳性60例(51.72%)。培养出阳性菌株80株,其中革兰阳性球菌占61.25%(松鼠葡萄球菌占28.75%,缓慢葡萄球菌占17.50%),革兰阳性杆菌(F-1棒杆菌)12.5%,革兰阴性杆菌25.00%(奇异变形杆菌为12.5%,铜绿假单胞菌为6.25%),说明B6-Co小鼠角膜混浊主要与革兰阳性球菌(松鼠和缓慢葡萄球菌)有关。结论:导致B6-Co小鼠角膜混浊的细菌主要为革兰阳性球菌(松鼠和缓慢葡萄球菌)。小鼠的角膜混浊可能与ENU引起的遗传基因变化导致的抗感染免疫力下降引起感染性角膜炎有关。Objective: To investigate the etiology of B6-Co mice, and the corneal opacification mutants and their offsprings, which were generated by ENU mutagenesis technology. Methods: Bacterial culture: 180 B6-Co mice varying from 1 day to 10 days were picked out as the experimental group. 88 normal B6 mice within the age from 1 day to 2 months were grouped as the control group. Biopsy scraping samples from the conjunctival sac and / or corneal lesions were cultured by blood plate and nutrient broth to identify the microorganisms. Results: In the B6 strain, the bacterial culture of conjunctival sac specimens was negative before they opened eyes (12-14 days). After they opened eyes, a total of 77 samples were isolated from the specimens, and the positive rate was 86.76%. Among them, Gram-pbsitive cocci accounted for 85.71%, Gram-positive bacterium rod for 13.00%. The detection rate of Staphylococci slow and Lira Micrococci was the highest (both 36.36%), followed by F-1 rods bacilli; In the B6-Co strain, among 116 specimens, the positive ones were 80, and the positive rate was 51.72%. Among them, Gram-positive cocci accounted for 61.25% (Squirrels Staphylococci were 28.75%, Staphylococci Slow were 17.50%), Gram-positive bacilli (F-1 Corynebacterium sp) accounted for 12.50%; Gram-negative bacilli were 25.00%(Proteus mirabilis was 12.50%, and Pseudomonas aernginosa was 6.25%). So B6-Co mouse keratitis may mainly caused by Gram-positive cocci (Staphylococci Sciuri and Staphylococci Slow).Conclusion: Gram-positive bacteria (Staphylococci aureus and slow squirrels grapes Cocci) are the major ones that caused bacterial keratitis of B6- Co strain. The corneal inflammation of B6-Co mice was related with the drop of anti-infection immunity which was caused by ENU-induced genetic changes.
