肝癌中COX-2蛋白与MMP-2蛋白的表达及其相关性研究

摘　　要：目的:探讨环氧化酶-2(COX-2)与基质金属蛋白酶-2(MMP-2)在肝细胞癌(HCC)中表达的相关性和临床意义。方法:应用免疫组化SP法检测47例HCC组织、33例癌旁组织COX-2蛋白和MMP-2蛋白的表达。结果:COX-2蛋白在HCC组织、癌旁组织中的阳性表达率分别为78.7%,30.3%(P<0.05),COX-2蛋白的表达与肿瘤分化程度、肝硬化、静脉浸润以及TNM分期相关(P<0.05);MMP-2蛋白在HCC组织、癌旁组织中的阳性表达率分别为68.1%,39.4%(P<0.05),MMP-2蛋白的表达与肿瘤大小、分化程度、静脉浸润、TNM分期相关(P<0.05)。COX-2与MMP-2同时阳性表达者为25例。结论:COX-2蛋白和MMP-2蛋白在HCC组织中表达上调,并呈正相关(P<0.01),证实COX-2和MMP-2在HCC发生发展过程中起重要作用。Objective：To investigate the expression of Cyclooxygenase-2; and matrix metalloproteinase-2 in HCC and the relationship between the expression and the clinical pathology parameters.Method：Expression of COX-2 and MMP-2 in the HCC tissues, paracancerous liver tissues respectively were detected by imunohistochemical method. Results：The COX-2 protein expression rate was 78.7% and 30.3%（P〈0.05） in the HCC tissues, paracancerous liver tissues respectively. The positive rate of COX-2 was correlated significantly with tumor cell differentiation, co-existing cirrhosis,vein invasion and TNM stage. The MMP-2 protein expression rate was 68.1% and 36.4%（P〈0.05） in the HCC tissues , paracancerous liver tissues respectively. The positive rate of MMP-2 was correlated significantly with tumor size,vein invasion, tumor cell differentiation and TNM stage. A close correlation was showed between the expression of COX-2 and MMP-2 （P〈0.01）. Conclusions： COX-2 and MMP-2 were over-expressed and significantly correlated in HCC tissues, which means COX-2 and MMP-2 might involve in the process of genesis and progression in HCC.
