摘  要:目的建立放射性脑损伤大鼠模型并观察P35、P25蛋白的表达情况。方法120只雄性SD大鼠随机分为单次全脑照射0、10、20、30 Gy 4个组。构建大鼠的放射性损伤模型,尼氏染色观察各组大鼠海马CAl区神经元的数目。组织匀浆法提取海马蛋白,Western blot法检测海马区P35、P25蛋白的表达。结果与0 Gy组比较,10 Gy组神经细胞死亡、P35及P25表达差异不明显(P>0.05);20、30 Gy组存活神经元减少,P35表达减少,P25表达增加(P<0.05)。结论放射性损伤可导致海马区神经元损伤,可能与P35和P25蛋白表达变化有关。Objective To establish a whole brain radiation injury model and observe the expression of P35 and P25 in hippocampal neurons. Methods 120 rats were randomly divided into 0, 10, 20, 30 Gy groups. The neurons of CA1 were detected by Nissl's staining. Hippocampus protein was extracted from these groups. Western blot was used to observe the expression of P35 and P25 in hippocampus. Results Compared with the 0 Gy group, the number of neurons in 10 Gy group showed no obvious change (P 〉 0.05). However, the number of neurons in 20 Gy and 30 Gy groups decreased obviously (P 〈 0.05 ). Compared with the 0 Gy group, the expression changes of P35 and P25 in 10 Gy group were not obvious ( P 〉 0.05 ). The P35 expression decreased while P25 expression increased in 20 Gy and 30 Gy groups ( P 〈 0.05). Conclusion The neural injury after radiation brain injury in hippocampns may be related with the expression changes of P35 and P25.
