摘  要:目的观察维生素C(VC)、维生素E(VE)及其联合作用对体外氧化损伤血管内皮细胞抗氧化作用的影响。方法采用体外细胞培养方法,将血管内皮细胞分为正常对照组,H2O2氧化损伤对照组,损伤加VC低、中、高浓度组(VC-L、M、H组),损伤加VE低、中、高浓度组(VE-L、M、H组),损伤加VC、VE联合低、中、高浓度组(VC-L+VE-L、VC-M+VE-M、VC-H+VE-H组)。分别将不同剂量的VC、VE加入到培养液中,预孵24h,再加入除菌后的H2O2进行损伤,终止培养后收集细胞,测定各组细胞抗氧化指标。结果与H2O2氧化损伤对照组比较,VC、VE、VC+VE各组均能增强其SOD、GSH-PX及CAT活性,降低LDH释放率,差别有统计学意义。与正常对照组相比较,(VC+VE)-M、H组差别无统计学意义。此外,(VC+VE)-L组与VC、VE-L差别无统计学意义,但(VC+VE)-M、H组SOD均高于VC、VE-M、H组,(VC+VE)-H组LDH释放率低于VC、VE-H组,差别有统计学意义。结论VC、VE能够提高氧化损伤血管内皮细胞的抗氧化能力,且二者联合作用优于单独作用。Objective To observe the protective effect of vitamin C (VC), vitamin E (VE) and their combination on vascular endothelial cells (VECs) following oxidative damage in vitro. Method Vascular endothelial cells cultured in vitro were divided into normal control, H2O2 oxidative control, (VC-L, M, H)+H2O2, (VE-L, M, H)+H2O2, and (VC+VE)-L, M, H+H2O2 groups. VECs were injured by H2O2 after they were incubated with different dosages of VC, VE or (VC +VE) for 24 h. The antioxidant indices of cell were measured. Results The activity of SOD, GSH-Px and CAT of cell in the (VC/VE-L, M, H)+H2O2 and (VC+VE)-L, M, H+H2O2 groups were significantly higher than those in the H2O2 oxidative control (P〈0.05 or P〈0.01), while the releasing percentage of LDH of cell was significantly lower than those in the H2O2 control. There was no significance in the (VC+VE)-M, H+H2O2 and normal control group. In addition, there was no significance in the effect of (VC+VE)-L+H2O2 and VC/VE-L+H2O2 group. But SOD of (VC+VE)-M,H+H2O2 groups was significantly higher than those in the VC/VE-M, H +H2O2 groups, and the releasing percentage of LDH of (VC+VE)-H+H2O2 groups was significantly lower than those in the VC/VE-H+H2O2 groups. Conclusion VC, VE can promote the antioxidative capability of VEC and prohibit H2O2 induced oxidative damage on VEC. The combined effect of VC and VE is better than the single effect.
