肺癌中GRIM-19 mRNA的表达及其临床意义

摘　　要：目的了解干扰素/维甲酸诱导凋亡相关基因(GRIM-19)在肺癌中的表达及其临床意义。方法RT-PCR检测40例肺癌患者的癌组织和癌旁组织中GRIM-19 mRNA表达水平,用Western blot法验证20例肺癌组织中GRIM-19蛋白表达情况,并分析GRIM-19 mRNA表达与临床病理参数之间的关系。结果肺癌组织中GRIM-19 mRNA表达水平低于癌旁组织,两组间差异有显著性(P<0.05),并且mRNA表达水平的降低伴随有蛋白表达水平的相应下降。GRIM-19 mRNA水平与临床病理分期之间有关联(P<0.05),Ⅰ～Ⅱ期Grim19 mRNA表达量显著高于Ⅲ～Ⅳ期中的表达,而与患者性别、年龄、组织分型、吸烟指数之间均无关联(P>0.05)。结论GRIM-19 mRNA的表达下降是肺癌发生过程中的重要事件,可能与肺癌的发生及进展有关。Objective To evaluate the expression of GRIM-19 mRNA in lung cancer and the relation to clinicophathological parameters. Methods The expression of GRIM-19 mRNA in 40 lung cancer tissues and the adjacent normal lung tissues was determined with RT-PCR methods. To further substantiate observations of GRIM-19 mRNA expression, the level of protein expression of the Griml9 in 20 samples was analyzed by Western blot analysis. Relationship between GRIM-19 mRNA expression and clinical and pathological parameters was analyzed. Results The expression of GRIM-19 mRNA in lung cancer tissue was significantly lower than that in normal tissue（P 〈 0.05 ）. A decreased GRIM-19 mRNA expression was corresponded to a similar effect on Grim-19 protein expression. The expression of GRIM-19 mRNA was not associated with the patients＇ sex, age, smoking condition and histology. However, its relationship with TNM staging was established （P 〈 0. 05 ）, and the relative amount of Grim19 mRNA expression of stage Ⅰ - Ⅱ were significantly higher than those of stage Ⅲ - Ⅳ. Conclusion Grim19 mRNA is down-regulated in tumor tissue. The lower expression of GRIM-19 mRNA may play a major role in lung cancer carcinogenesis, which suggests that may have very intimate association with the development of lung cancer.
