摘  要:目的:研究ADAM28反义核酸(AS-ODN)对人牙囊细胞(HDFC)增殖、分化、凋亡等生物学特性的影响。方法:采用细胞培养、基因转染、四唑盐(MTT)比色法、酶动力学方法和流式细胞术(FCM)检测ADAM28反义核酸转染HDFC后对细胞生物学特性的影响。结果:ADAM28 AS-ODN组HDFC的增殖活性、增殖指数、碱性磷酸酶(ALP)分泌活性显著降低,凋亡细胞百分比明显上升。结论:ADAM28反义核酸可显著抑制HDFC的增殖和ALP的活性,显著诱导HDFC的凋亡并影响细胞周期的变化。Objective: To study the effects of ADAM28 antisense oligodeoxynucleotide (AS -ODN) on biological char- acteristics of human dental follicle cells (HDFC) such as proliferation, differentiation and apoptosis. Methods: Cell culture, gene transfection, MTT chromatometry, enzyme dynamics, and flow cytometry (FCM) techniques were used to detect the effects of ADAM28 AS - ODN on biological characteristics of HDFC after transfected into HDFC. Results : In ADAM28 AS -ODN group, proliferation activity, proliferation index and alkaline phosphatase(ALP) secretion activity of HDFC decreased significantly, and percentage of cell apoptosis were up - regulated obviously. Conclusion: ADAM28 AS - ODN could inhibit the proliferation and ALP activity of HDFC significantly, induce apoptosis of HDFC and influence the changes of cell cycle significantly.
