摘 要:研究发现与IL-1诱导反应相关的APP启动子-488/-303片段,可与SH-SY5Y细胞核抽提物中的核蛋白发生结合作用,并且在IL-Ⅰ刺激前后有量的变化,IL-Ⅰ刺激后其结合作用明显加强.冷竞争实验表明,此结合作用具有特异性.同时发现APP启动子-488/-303片段中的AP-Ⅰ作用位点及SH-SY5Y细胞核抽提物中的AP-Ⅰ作用因子可能与此结合反应无关,而一个90kD蛋白质与此结合反应有关.Previous studies have demonstrated that the expression of APP gene was increased in the SH SY5Y cell by induced of IL Ⅰ.Gel shift data indicate that the fragment of APP promoter -488/-303,which has been implicated as potential response element in the induction of APP transcription by IL Ⅰ,may interact with nuclear protein extracted from SH SY5Y cell.IL Ⅰ induction cause some modulation in amount of bound protein.Gel shift self competition assay showed that the binding is specific and expression of APP gene is not regulated efficiently by AP Ⅰ factor binding AP Ⅰ site.Instead,Southwestern blot result showed that a 90 kD protein may play a more important role.The data suggest that a 90 kD protein involve the regulation of APP gene expression in the SH SY5Y cell by induced of IL Ⅰ .
