摘 要:目的采用高效液相色谱(HPLC)法分离纯化促黄体生成激素释放激素(LHRH)拮抗类似物。方法制备型色谱柱为Dynamax-300A型C18柱(250mm×4.14mm,5μm),分析型色谱柱为Nucleosil型C18柱(250mm×5mm,5μm);流动相A为含0.1%三氟醋酸和3%乙腈的水溶液(pH=2),流动相B为含70%乙腈的水溶液;流速为40mL/min(用于分离)和1.2mL/min(用于分析);检测波长为220nm。结果LHRH拮抗类似物纯品的产率可达22%~52%,纯度高达97%~99%,分离度大于1.5。结论HPLC法分离纯化的LHRH拮抗类似物纯品产率大,纯度高,且该法操作简便、快捷。Objective To inquire into the separation and purification of luneinizing hormone releasing hormone (LHRH) antagonists by HPLC. Methods HPLC was used. The preparative type column (Dynamax-300A C18, 250 mm ×4. 14 mm, 5 μm) and analytical type column (Nucleosil C18, 250mm×5 mm, 5 μm);the aqueous solution of 1% trifluoroacetic acid and 3% acetonile as the mobile phase A(pH=2), and the aqueous solution of 70% acetonilc as the mobile phase B; the flow rate was 40.0 mL/min (for separation) and 1.2 mL/min (for analysis), the detecting wavelength was 220 nm. Results The product yield of purification was 22%-5267o and the purity was 97%- 99%, the resolution was large than 1.5. Conclusion The method is simple and fast. Therefore LHRH antagonists can be well purified by the way of HPLC.
