摘 要:从血液涂片检查为牛瑟氏泰勒虫阳性的病牛血液中提取总RNA,通过RT-PCR技术扩增出牛瑟氏泰勒虫HSP70基因,将其重组到pMD-18TSimple载体后进行克隆、序列测定及分析。结果表明该片段长1 966 bp,编码620个氨基酸残基,将该基因片段序列与GenBank中13种已知梨形虫的相应序列进行比较分析,牛瑟氏泰勒虫吉林分离株与已报道的牛瑟氏泰勒虫亲缘关系最近,其次是环形泰勒虫,与马巴贝斯虫亲缘关系较远。Total RNA was extracted from the diseased cattle in which Theileria were found in the blood smears. The HSP70 gene fragment of cattle T. sergenti was obtained by RT-PCR and cloned into pMD-18T simple vector for sequencing. The result indicated that the cDNA sequence was 1966 bp in length, coding 620 amino acid residues. The complete gene sequence was compared with those of 13 Piroplasma species available in GenBank. The phylogenetic tree of HSP70 genes indicated that Jilin strain of cattle T. sergenti was closest to T. sergenti, less to T. annulata, but farthest to Babesia equi.
