恶性胸水淋巴细胞体内、外培养抗肿瘤的研究

摘　　要：对恶性胸水肿瘤浸润淋巴细胞(TIL)体内、体外培养的抗肿瘤作用进行了比较研究。结果显示,在恶性胸水患者胸腔内注入基因重组白介素-2(rIL-2)及基因重组肿瘤坏死因子(rTNF-α)能起到体内培养 TIL 的作用,且 TIL 扩增倍数高;对自体肿瘤细胞(ATC)的细胞毒作用亦明显升高,且高于体外培养(P<0.01)。临床治疗11例恶性胸水患者均取得较好效果,8例胸水中 ATC 检测转阴,较体外培养法有较大的优越性。In order to obtain a simple and effective therapy of malignant pleural effusion,we made a comparative study on the antitumor effects of tumor infiltrating lymphocytes(TIL)in vivo and in vitro.The data showed that TIL can be induced in vivo by intrapleural instillation of rIL-2 and rTNF-x in patients with malignant pleurisy,its cytotoxicity against the autologous tumor cells was significantly higher than that in vitro(P<0.001).In addition,the malignant pleural effusion itself which served as a cell culture medium in vivo had the cytotoxicity effect.Good results were observed in all 11 patients studied.The general condition improved in all patients and the ATC test of pleural effusion became negative in 10 of them.We believe that it is a new approach to the adoptive immunotherapy of cancer with TIL,which can be served as a substitute for the routine therapies of this disease.
