dNTP库与细胞转化间关系的研究

摘　　要：采用反相ＨＰＬＣ法定量测定了ＢＡＬＢ／ｃ３Ｔ３细胞经烷化突变剂甲基丙烯酸环氧丙酯（ＧＭＡ）和Ｎ－甲基－Ｎ′－硝基－Ｎ－亚硝基胍（ＭＮＮＧ）转化处理后胞内脱氧核糖核苷三磷酸含量（ｄＮＴＰ库）的变化及最终形成的Ⅲ型转化细胞中的ｄＮＴＰ库，发现转化处理后ｄＮＴＰ库的变化规律相似：均表现为脱氧嘌呤核苷三磷酸库与脱氧嘧啶核苷三磷酸库间差距明显扩大；转化细胞中ｄＮＴＰ库也明显异于正常细胞，提示ｄＮＴＰ库可能在细胞转化进程中起着重要作用。Deoxyribonucleoside triphosphate(dNTP) pools were measured in normal BALB/c3T3 cells,transformation treated cells and transformed cells with reverse phase HPLC.The fluctuation of dNTP pools was similiar after cells were treated with alkylating mutagens glycidyl nethacrylate(GMA) and N methyl N′ nitro N nitrosoguanidine(MNNG).The gap between (dGTP+dATP) pools and (dTTP+dCTP) pools was greatly intensified.The measurements also indicated that the dNTP pools in transformed cells were quite different from those in normal cells.The results suggest that dNTP pools may play an important role in cell transformation.
