摘  要:目的探讨烟草浸提液对钛板表面牙龈成纤维细胞分泌胶原的影响。方法取细胞悬浮液接种于24孔板内钛板表面,孵育24h后,更换含0、0.01、0.1、1、5、10、25g/L烟草浸提液和0.03、0.15、0.3g/L尼古丁(10种浓度)的培养液。继续孵育48h后,检测细胞培养上清液中羟脯氨酸含量。结果随着烟草浸提液及尼古丁浓度的增加,细胞培养上清液中羟脯氨酸的含量逐渐减少。结论烟草浸提液及尼古丁可抑制牙龈成纤维细胞胶原的产生,提示烟草在破坏牙种植体"袖口"中起重要作用。Objective To evaluate the effects of smokeless tobacco extract (ST) on collagen synthesis of human gingival fibroblasts(HGFs) to titanium in vitro. Methods HGFs were implanted on the surface of titanium for 24 h and then cultured in DMEM containing 0,0.01,0. 1, 1,5, 10,25 g/L ST and 0.03,0.15,0.3 g/L nicotine respectively. After 2 d, the cell culture supernatants were collected. Hydroxyproline chromatometry was performed to detect the level of collagen. Results The content of Hydroxyproline chromatometry of cell culture supernatants was reduced when the concentration of ST and nicotine increased. The synthesis of collagen of HGFs was dose-dependently inhibited. Conclusion ST and nicotine reduced the synthesis of collagen of HGFs in a dose-dependent manner, suggesting ST may have pathological role on implant-gingiva interface.
