摘 要:目的探讨电离辐射对胶质瘤C6细胞垂体瘤转化基因(pituitary tumor transforming gene,PTTG)表达的影响。方法胶质瘤C6细胞分为4组(n=6):A组(空白对照组)、B组(2 Gy剂量X射线组)、C组(4 Gy剂量X射线组)、D组(6 Gy剂量X射线组),X射线照射后12 h,逆转录聚合酶链反应(RT-PCR)检测PTTG mRNA的表达,免疫细胞化学检测其蛋白表达。结果PTTG mRNA在X射线处理组均降低,随着X射线剂量的增加,其表达降低越明显,A、B、C、D 4组的表达分别为1.910±0.141、1.398±0.157、1.010±0.133、0.743±0.092,各组间差异均有统计学意义(P<0.01)。PTTG蛋白积分光密度(integrated optical density,IOD)在X射线处理组均降低,随着X射线剂量的增加,其表达降低越明显,A、B、C、D 4组的IOD分别为1.840±0.089、1.395±0.139、0.893±0.125、0.597±0.104,各组间差异均有统计学意义(P<0.01)。结论电离辐射可以剂量依赖的方式降低胶质瘤C6细胞PTTG mRNA及其蛋白的表达。Objective To investigate the effect of ionizing radiation on the expression of pituitary tumor transforming gene(PTTG) in glioma C6 cells.Methods Glioma C6 cells were divided into 4 groups(n=6): group A(control group),group B(2 Gy doses of X-ray group),group C(4 Gy doses of X-ray group),group D(6 Gy doses of X-ray group).The expression of PTTG mRNA was detected by reverse transcription polymerase chain reaction(RT-PCR) 12 hours after X-ray irradiation and the expression of PTTG protein was detected by immunostaining assay using streptavidin-peroxidase(SP) method.Results The expression of PTTG mRNA decreased in X-ray treatment groups,which was positively correlated with the increase of X-ray doses.The expression of PTTG mRNA in groups A,B,C and D were 1.910±0.141,1.398±0.157,1.010±0.133,0.743±0.092,respectively.There were significant differences between these groups(P〈0.01).The expression of PTTG protein decreased in X-ray treatment groups,which was also positively correlated with the increase of X-ray doses.The integrated optical density(IOD) of PTTG protein in groups A,B,C and D were 1.840±0.089,1.395±0.139,0.893±0.125,0.597±0.104,respectively.There were significant differences between these groups(P〈0.01).Conclusion Ionizing radiation can decrease the expression of PTTG mRNA and the protein in glioma C6 cells in a dose-response relationship.
