猪Sirtuin3基因的克隆与进化分析

摘　　要：从猪十二指肠肠道组织提取总RNA,采用RT-PCR方法克隆了猪Sirtu in 3基因,并与GenBank中的9个其他物种的核酸序列进行了同源性分析,采用邻接法构建了分子系统进化树。结果表明克隆的猪Sirtu in 3基因长999 bp,编码332个氨基酸,前21个为信号肽;猪Sirtu in 3与牛、人的同源性最高,分别为86.3%、79.6%,分子进化树也表明猪的Sirtu in 3与牛、人的亲缘关系最近。Total RNA was isolated from porcine duodenal perforation tissue. The Sirtuin3 cDNA was amplified by RT -PCR and then cloned and sequenced. The nucleotide sequence was compared with the homologous genes in other species retrieved from the Gen- Bank, and finally the phylogenetic tree was constructed by NJ method. The results indicated that the cloned porcine Sirtuin 3 gene was 999 bp in length, the deduced amino acids had 332, and the signal peptide of predicted amino acid sequence was the 1 ～21st amino acid. Homology analysis showed that the sequence shared 86.3% and 79.6% homology with those of cow and human respectively. Phylogenetic analysis based on the nucleotide sequence from the Sirtuin3 gene showed that pig had a closer phylogenetic relation with cow and human.
