摘  要:目的观察卡介苗(BCG)及其缓释剂型BCG-明胶和BCG-泊洛沙姆407(Poloxamer407,P407)对荷瘤大鼠肿瘤细胞与胸腺细胞增殖、凋亡的影响。方法在大鼠皮下建立肿瘤模型,3d后在肿瘤结节内分别直接注射BCG(BCG组)、BCG-明胶(明胶组)、BCG-P407(P407组),对照组注射PBS,每组10只大鼠。在接种后7和14d取肿瘤结节行常规病理观察,取胸腺行流式细胞术(FCM)检测。结果肿瘤病理观察,BCG组和P407组可见肿瘤坏死和类细胞浸润,以P407组为重。FCM检测结果表明,单纯注射BCG组,在14d时显示了胸腺细胞的高凋亡率,胸腺细胞的增殖活性从7~14d呈现下降趋势;P407组,在14d时显示了胸腺细胞的低凋亡率,胸腺细胞的增殖活性从7、14d一直处于比较高的状态;明胶组,在7、14d时均显示了胸腺细胞的高凋亡率,胸腺细胞的增殖活性在7和14d无明显区别。结论P407与BCG联合应用,将有助于提高BCG的肿瘤治疗和预防作用。Objective To observe the effect BCG, BCG-gelatin and BCG-Poloxamer 407 (BCG-P407) on proliferation and apoptosis of thymoeyte of tumor-bearing rat. Methods Tumor model was made by subcutaneously injected tumor. 3 days later, BCG, BCG-gelatin and BCG-P407 were respectively injected into tumor tubercles, and on the 7th and 14th day, the tubercles were made pathologic examination, and thymus was made FCM test. Results Apoptosis rate of thymocyte was high on the 14th day and the proliferation of thymocyte declined gradually from 7th to 14th day in BCG group. Apoptosis rate of thymocyte was low on the 14th day and the proliferation of thymocyte was always in high condition from 7th to 14th day in BCG-P407 group. Apoptosis rate of thymocyte was high on the 7th and 14th day and the proliferation of thymoeyte had no difference between the 7th and 14th day in BCG-gelatin group. Conclusions P407 could contribute to improving BCG's therapeutic and preventative effect on tumor.
