摘  要:[目的]探讨cox-2抑制剂赛来昔布(Celecoxib)对骨肉瘤类肿瘤干细胞裸鼠移植瘤生长及微血管生成的影响。[方法]无血清培养法从骨肉瘤细胞株MG-63中分离出类肿瘤干细胞建立裸鼠移植瘤模型。30只成瘤裸鼠随机分Celecoxib组和对照组,Celecoxib:25 mg/(kg.d),用药15 d,第27 d处死裸鼠,观察肿瘤体积、抑瘤率,免疫组化技术检测VEGF表达及CD34标记的MVD值。[结果]分离的骨肉瘤类肿瘤干细胞有致瘤性,可以建立动物模型。Celecoxib抑瘤率为23.2%,Celecoxib组裸鼠移植瘤的体积、VEGF的表达、MVD值均显著低于对照组(P<0.05)。[结论]骨肉瘤类肿瘤干细胞可以建立裸鼠骨肉瘤移植瘤模型。Celecoxib可以抑制肿瘤生长,减少移植瘤组织VEGF的表达,减少微血管生成,具有抗血管生成作用。[ Objective] To explore the effects of celecoxib on growth and angiogenesis of the xenografted human osteosarcoma stem - like cells in nude mice. [ nethod] Human osteosarcoma stem - like cells were isolated from human osteosarcoma cell line MG - 63 with serum - free culturing techniques. The xenografted models of human osteosarcoma stem - like cells were successfully established using subcutaneous injection of human osteosarcoma stem - like cell in 30 nude mice and randomly divided into the control and celecoxib groups. Oral administration was carried out in both groups , with 25 mg/kg/d of celecoxib in the celecoxib group and the same volume of deionized water in the controls. This treatment lasted for 15 days and all mice were sacrificed 27 days after to investigate the tumor volumes and tumor inhibitory rate. VEGF expression and CD34 tagged microvessel density (MVD) value was detected by immunohistochemical staining. [ Result]The isolated human osteosarcoma stem - like cell exhibited tumorigenesis and the animal model was established at 15 days of administration. The tumor inhibitory rate was 23.2%, and the tumor volumes, VEGF expression and MVD values were significantly lower than those in control group ( P 〈 0. 05) . [ Conclusion ] Human osteosarcoma stem - like cells can be used to establish xenografted osteosarcoma model in nude mice. Celecoxib showed a significantly inhibitive effects on growth, VEGF expression and angiogenesis of xenografted osteosarcoma.
