摘  要:目的探讨咪唑克生(Ida)对实验性自身免疫性脑脊髓炎(EAE)Lewis大鼠脊髓内胶质纤维酸性蛋白(GFAP)蛋白及其mRNA表达的影响。方法将30只雌性Lewis大鼠分为三组:正常对照组、EAE组和Ida组。以豚鼠脊髓匀浆(GPSCH)诱发制备大鼠EAE模型,并于给予GPSCH当日开始分别腹腔注射10mg/kgIda(Ida组)和生理盐水(EAE组和对照组)连续10d,观察EAE症状并评分。用免疫组化法和逆转录-聚合酶链反应(RT-PCR)法分别检测GFAP蛋白及mRNA在脊髓中的表达。结果10d后Ida组大鼠的临床症状和病理学改变减轻(<0.05)。脊髓组织中GFAP蛋白及其mRNA的表达,EAE模型组显著高于对照组,Ida组较EAE组显著增高,差异有统计学意义(<0.05)。结论Ida可促进GFAP在EAE大鼠脊髓中的表达,可能是其保护EAE大鼠的部分作用机制。Objective To study the effects of idazoxan(Ida) on the expression of GFAP and its mRNA in in spinal cords of Lewis rat with experimental autoimmune encephalomyelitis (EAE). Methods Thirty female Lewis rats were randomly divided into three groups: the control group, EAE group and idazoxan(Ida)-treated group. In the experiment,the rat models of EAE were established by induction with Guinea pigs spinal cord homogenate(GPSCH). Idazoxan were injected intraperitoneally at 0-10 days after immunization successively in Ida-treated group; meanwhile, saline were injected intraperitoneally at 0-10 days after immunization successively in EAE group and control group. And then clinical scores were observed. The expression of GFAP protein and mRNA in spinal cord were detected by immunohis- tochemistry and reverse transcription polymerase chain reaction (RT-PCR), respectively. Results Ida could alleviate its clinical manifestations and histological changes(P〈0.05,respectively). Immunohistochemistry and RT-PCR showed that the protein and mRNA expression of GFAP in Ida group were significantly higher than those of control group (P 〈 0.05, respectively). While those of DXM group were significantly higher than those of EAE group (P 〈 0.05, respectively). Conclusion The beneficial effect of Ida on EAE may be in part due to their direct promotive effect on GFAP generation in spinal cord.
