摘 要:目的探讨金黄色葡萄球菌DNA(S.aureus DNA)抗肿瘤免疫的效果及其免疫机制。方法纯化提取S.aureus DNA,建立荷瘤裸鼠模型,将24只荷瘤裸鼠随机分为TES组、BCG组、DNA组,每组8只。于接种A549细胞株后第15天,TES组、BCG组、DNA组分别腹腔注射TES液、BCG液、DNA液各0.3 mL,隔2 d 1次,共8次。采用双抗体夹心酶联免疫检测法检测3组荷瘤裸鼠血清IFN-γ、TNF-α水平,并进行NK、M细胞杀伤活性、S.au-reus DNA体内抑制率、不同肿瘤细胞抑制率的测定。结果DNA、BCG、TES3组间NK、M细胞杀伤活性比较差异均无统计学意义(P均>0.05)。DNA、BCG2组瘤重均明显低于TES组(P均<0.05)。SPC-A-1、MGC803 100mg/L S.aureus DNA抑制率均明显高于A549(P均<0.05),A549、MGC803 50 mg/L S.aureus DNA抑制率均明显低于SPC-A-1(P均<0.05)。TES、BCG2组血清IFN-γ、TNF-α水平均低于DNA组(P均<0.01)。结论S.aureus DNA具有抗肿瘤免疫特性。Objective To explore the antitumor effects of staphylococcus aureus DNA (S. aureus DNA)and its mechanism. Methods S. aureus DNA was extracted and purified, and the tumor-bearing models of nude mice were established, 24 tumor-bearing nude mice were randomly divided into TES,BCG and DNA groups and each group had 8 mice. After Vaccination A549 cell line 15 days,the mice in TES group,BCG group and DNA group were injected peritoneally with TES,BCG and DNA solution. Each tumor-bearing nude mice was injected 0.3 mL,once every 2 days, a total of 8 times. The serum levels of IFN-γ, and TNF-α were detected by enzyme-linked immunosorbent assay,and killing activity of NK, Mφ, cell of three groups tumor-bearing nude mice, the suppression rate of tumor in vivo and different tumor cells in vitro were detected. Results NK, Mφ cell killing activity were compared in DNA group, BCG group, and TES 3 group and no statistical significance was found (all P〉0.05). The serum levels of IFN-γ and TNF-α as well as tumor weight were significantly lower than those in TES group (all P 〈 0. 05). SPC-A-1, MGC803 100 mg/L S. aureus DNA suppression rate were significantly higher than A549 (all P〈2 0.05), A549,MGC803 50 mg/L S. aureus DNA suppression rate were significantly lower than SPC-A-1 (all P〈0.05). TES, BCG 2 group serum levels of IFN-γ, TNF-α were lower than DNA group (all P〈0.01). Conclusion S. aureus DNA has the characteristics of antitumor immune.
