摘  要:本研究建立一种以过氧草酸酯为化学发光体系的新型毛细管电泳化学发光系统,并将该系统应用于氨基酸的分离测定。该系统设计了新型的电极模式与反应混和模式以获得高的信号稳定性与检测灵敏度。以一端与高压电源负极相连的铂丝连接到不锈钢注射针头为接地电极,电泳毛细管穿过注射针头,且其末端到达注射针头的出口,电泳毛细宇航局末端与化学发光试剂在反应池中以相对的方向靠近。实验结果表明,该系统稳定性好、分离效率高、灵敏度高、精密度好,对天冬氨酸与亮氨酸的检出限分别为2.0和1.1nmol/L。峰高及迁移时间的相对标准偏差分别为2.3%~3.8%和1.2%~1.5%。A peroxyoxalate-based chemiluminescence(CL) detection following capillary electrophoresis (CE) separations for assaying amino acids is described. To achieve high signal stability and sensivity, a new interface was designed including a new mixing mode and a new grounding electrode mode. A platinum wire-connected cathode of a high voltage source was put onto a stainless steel needle tube, which was used as grounding electrode. The CE capillary was inserted into the stainless steel needle tube and the capillary outlet was put on the outlet of the needle tube. The CE capillary and two reagent capillaries used for delivering chemiluminescence reagent were fixed oppositely to a reaction cell. Amino acids fluorescently tagged with dansyl chloride and naphthalene-2,3-dicarboxaldehyde(NDA) were used for the study. Results show this CE-CL system has good stability, resolution, sensitivity and reproducibility. The detection limits were 1.1 nmol/L for dansyl-leucine (Leu) and 2.0 nmol/L for dansyl-aspartic acid (Asp). The relative standard deviations of peak height and migration time were 2.3% -3.8% and 1.2% - 1.5%, respectively.
