摘　　要：目的研究大鼠间充质干细胞(MSCs)对BALB/c小鼠淋巴细胞增殖和CD4+CD25+调节性T细胞亚群的变化。方法分离、扩增SD大鼠MSCs,流式细胞术检测表面标志以鉴定。取第5代MSCs与分离的小鼠淋巴细胞在刀豆蛋白(ConA)刺激下共培养5d,流式细胞仪测定细胞增殖,CD4+CD25+调节性T细胞亚群比例的变化。结果当淋巴细胞:MSCs在1∶10及1∶50时均可抑制ConA刺激下可抑制小鼠淋巴细胞增殖,淋巴细胞:MSCs在1∶10时CD4+CD25+调节性T细胞亚群比例明显升高。Objective To study the effects of rat bone marrow - derived mesenchymal stem cells （MSCs） on BALB/c mice T lymphocytes proliferation and on the change of CD4＋ CD25 ＋ regulatory T cell. Methods MSCs were isolated from SD rat, euhured in vitro, purified and then identified by examining surface marker with FCM. BALB/e mice Lymphocytes were co - cultured for 5 days. Proliferation of T lymphocyte and change of CD4^＋ CD25^＋ regulatory T cell were measured with FCM. Results When MSC and lymphocyte mixed at the ratio of 1 ： 10 and 1 ： 50, rat MSCs inhibited ConA - induced T lymphocyte proliferation. When MSC and lymphocyte were mixed at the ratio of 1： 10, the proportion of CD4^＋ CD25^＋ regulatory T cell increased. Conclusion Rat MSC can inhibit the immune response of xenogenic T lymphocytes.
