摘 要:目的探讨川芎嗪(TMP)对腹腔间隔室综合征(ACS)致肝损伤的保护作用机制。方法将60只新西兰白兔随机分成对照组、ACS2h和4h组、TMP干预2h和4h组五组,每组各12只。制备ACS兔实验动物模型;TMP干预组在灌注氮气前1h给予TMP处理,实验结束后处死动物,检测相关指标。结果ACS兔实验动物模型随着血内毒素浓度升高,血ALT值升高,线粒体Na^+-K^+-ATP酶、Mg^+-Ca^+-酶活力、肝脏GSH含量降低。TMP干预2h组较ACS2h组血清内毒素和ALT值下降(t=6.96,P〈0.05;t=1.24,P:0.23),线粒体Na^+-K^+-ATP酶、Mg^2+-Ca^2+-ATP酶活力、肝脏GSH含量升高(P〈0.05)。但TMP干预4h组较ACS4h组血清内毒素和ALT值显著升高,线粒体Na^+-K^+-ATP酶活力、Mg^2+Ca^2+-酶活力、肝脏GSH含量显著降低(P均〈0.05)。结论TMP干预早期能保护肝细胞线粒体功能,降低血内毒素浓度;但如ACS不能得到有效缓解,随着时间的延长,TMP的保护作用丧失。Objective To study the protective effect and probable mechanism of tetrameth ylpyrazine{ TMP) on liver injury with Abdominal Compartment Syndrome. Methods Sixty New Zealand white rabbits were randomly divided into five groups: normal control group(C), ACS 2 hours group (AC2) , ACS 4 hours group ( AC4 ) , TMP treatment 2 hours group ( T2 ) and TMP treatment 4 hours group( T4), 12 rabbits being in each group. The ACS model rabbits were produced by inflation of nitrogen into the rabbits. In TMP treatment group were treated with TMP before making ACS model for 1 hour. The rabbits were killed after experiment. The GSH content in the liver, the mitochondrial activity of ATPase , ALT and endotoxin levels were measured. Results In ACS model groups,more increasable the endotoxin levels was, more significently the level of the ALT increase, but more significently the GSH content in the liver, the mitochondrial activity of Na^+ - K^+ - ATPase and Mg^2+ - Ca^2+ - ATPase decrease. Compared with T2 group,The GSH content in the liver, the mitochondrial activity of Na^+- K^+ - ATPase, Mg^2+ - Ca^2+ - ATPasewere decreased( P 〈 0.05 ), but the ALT ( t = 1.24, P = 0.23 ) and endotoxin levels ( t = 6. 96, P 〈 0.05 ) were elevated significantly in AC2 group. But, the ALT and endotoxin levels were elevated significantly,the GSH content in the liver, the mitochondrial activity of Na^+ - K^+ - ATPase, Mg^2+ - Ca^2+ - ATPase were decreased significantly in T4 group ( all P 〈 0.05 ). Conelusion TMP may protect liver from injury in rabbit with early ACS, due to stabilizing hepatic mitochondrial redox status and energy level,decreasing endotoxin levels in blood, however, if ACS cannot be subdued effectively, experimental animals'conditions can be aggravated by TMP as time passes by.
