摘 要:目的:探讨抑制核转录因子κB(NF-κB)表达对宫颈癌HeLa细胞放疗敏感性的影响。方法:通过对HeLa细胞加入NF-κB的抑制剂100μmol/LPDTC或2μmol/L PS-341,均作用2 h来抑制NF-κB的表达。将HeLa细胞分为PDTC组、PS-341组及对照组加入等量生理盐水。各组均给予直线加速器照射,剂量分别为0、2、4和6 Gy。用蛋白质印迹法来观察细胞核内NF-κB含量的变化,用TUNEL法来检测细胞的凋亡,用MTT法检测细胞增殖。结果:辐射可以使HeLa细胞内NF-κB的表达增多,PDTC及PS-341均可以抑制由射线介导NF-κB的增加。在0 Gy条件下对照组与PDTC组及PS-341组HeLa细胞的凋亡指数分别为3%、12%和15%,两实验组的凋亡指数均高于对照组,P<0.01;6 Gy条件下实验组细胞的凋亡指数都高于同条件下对照组,P<0.01。结论:在宫颈癌HeLa细胞中,PDTC及PS-341均可抑制NF-κB的表达从而增加了由射线介导的细胞凋亡,增加HeLa细胞的放疗敏感性。OBJECTIVE:To explore effect on the radiation sensitivity of HeLa ceils by inhibiting expression of NF-κB. METHODS: Inhibition of NF-κB activation in HeLa cells was performed by treatment with 100 μmol/L PDTC or 2 μmol/L PS-341 for 2 h. Cells were irradiated at 0, 2, 4 and 6 Gy with or without inhibitor of NF-κB. The expression of NF-κB in nuclear was determined by western blot, and apoptosis was evaluated using the TUNEL assay. Cell proliferation was evaluated using MTT assay. RESULTS: NF-κB activation was induced by radiation and inhibited by PDTC and PS-341. At 0 Gy, the apoptosis index of control group, PDTC group and PS-341 group was 3%, 12% and 15% respectively; the AI in the experimental groups were higher than that of control group (P〈 0.01). Meanwhile, the same result can be found at 6 Gy (P〈0.01). CONCLUTION: Inhibition of the expression of NF-%B increases radiation induced apoptosis and enhances radiation sensitivity in uterine cervix cancer HeLa cells.
