子宫肌瘤组织Transgelin基因表达及其临床意义的研究

摘　　要：目的:检测Transgelin基因mRNA及蛋白在子宫肌瘤中的表达,探讨Transgelin基因与子宫肌瘤发生、发展的关系。方法:采用逆转录-聚合酶链反应(RT-PCR)及蛋白质印迹法,检测30例子宫肌瘤组织、30例子宫肌瘤包膜外2 cm子宫肌层组织和10例正常子宫肌层组织中Transgelin基因mRNA及蛋白的表达。结果:Transgelin基因mRNA在子宫肌瘤组织中阳性表达率及相对含量分别为23.3%和50.57±20.16,均低于子宫肌瘤包膜外2 cm子宫肌层组织的100.0%和84.62±6.12以及正常子宫肌层组织的100.0%和85.99±6.99,P<0.001。子宫肌瘤包膜外2 cm子宫肌层组织和正常子宫肌层组织中Transgelin基因mRNA的相对含量差异无统计学意义,P>0.05。Transgelin蛋白在子宫肌瘤组织中阳性表达率及相对含量分别为16.7%和23.46±16.54,均低于子宫肌瘤包膜外2 cm子宫肌层组织的100.0%和61.08±8.73以及正常子宫肌层组织的100.0%和67.19±7.62,P<0.001。子宫肌瘤包膜外2 cm子宫肌层组织和正常子宫肌层组织中Transgelin蛋白的相对含量差异无统计学意义,P>0.05。结论:Transgelin基因可能是子宫肌瘤发生过程中的抑制因素,与子宫肌瘤的发生、发展可能有关。OBJECTIVE： To investigate the expression of Transgelin in uterine myoma tissues and to reveal its role in genesis and progression of uterine myoma, soas to provide a theoretical basis for new conservative means to treat uterine myoma. METHODS： Reverse transcription polymerase chain reaction（RTPCR） and Western Blot were used to assess the expression of Transgelin mRNA and protein in 30 cases of uterine myoma tis-ues, 30 cases of normal myometrium tissues which were over 2 em far away the wrapping mambrane of myoma （as control） and 10 cases of normal myometrium tissues without myoma. RESULTS：The positive expression and relative content of Transgelin mRNA in uterin myoma tissues （23.3%, 50. 57±20. 16 ） were statistically lower than those in the control （100.0%, 84. 62±6.12） and the normal myometrium tissues without myoma （100.0%, 85.99±.6.99）, P〈0. 001. No significant differences between those in the control and in the normal myometrium tissues without myoma （P〉 0.05）. The positive expression and relative content of Transgelin protein in myometrium tissues （16.7%, 23. 46±16. 54） were statistically lower than those in the control （100.0%, 61.08±8.73） and the normal myometrium tissues without myoma （100.0%, 67.19±7.62）, P〈0. 001. No significant differences between those in the control and in the normal myometrium tissues without myoma （P〉0.05）. CONCLUSION： Transgelin may play a suppression role in genesis of uterine myoma, and be relative to the genesis, progression and prognosis.
