摘 要:为探讨牛血清中蛋白组分对细胞培养的影响,应用凝胶过滤层析对不同牛血清进行初步组分分析,并应用MTT比色法衡量不同牛血清的促细胞生长作用。结果表明,蛋白组分大小相对较为一致的新生牛血清、商品血清1、2、5(一个波峰)与蛋白组分差异较大的成年牛血清、商品血清34、(两个或者两个以上波峰)的促细胞生长作用相比较差异显著(P<0.05)。因此,牛血清中蛋白组分的不同,对细胞的促生长作用有很大影响。In order to investigate the cell culture which was influenced by protein compositions of bovine serum,this work was to make a preliminary anlysis of the protein compositions of bovine serum by the method of gel filtration chromatography(GFC) and to measure the effects of promoting the growth and proliferation of cell that using different bovine serum by the MTT assay. The results showed that the serum contains the relatively uniform protein compositions which were one peak through the GFC(newborn bovine serum, commercial serum 1, 2, 5) had significant differences compared with the serum contains great difference protein compositions which were two or more than two peaks through the GFC(adult bovine serum, commercial serum 3, 4) (P〈0.05). Therefore, the different compositions of bovine serum can result in different effects on promoting the growth and proliferation of cells.
