摘  要:目的探讨缺锌对大鼠胚胎心脏的发育毒性及其作用阈剂量和剂量-效应关系。方法50只健康清洁级SD雌性大鼠,随机分成5组:1极低锌组,2低锌组,3中等低锌组,4边缘低锌组,5常锌组,分别饲以不同含锌量的饲料。锌耗竭性饲养25天后交配,孕第19天时活杀取胚鼠,通过切片观察胚胎心脏畸形情况。结果各低锌组血锌值和血清碱性磷酸酶活性显著低于常锌组(血锌比较,t值分别为49.44、27.92、21.10、8.14,均P〈0.05;血清碱性磷酸酶比较,t值分别为86.79、52.37、38.32、13.35,均P〈0.05)。同等条件下血清碱性磷酸酶活性变化更为明显。极低锌组、低锌组和中等低锌组与常锌组相比,胚心畸形率均明显增高(,分别为35.62、16.93、5.91,均P〈0.05);边缘低锌组与常锌组间比较则无显著性差异(X2=0.00,P〉0.05)。胚心畸形主要表现为发育延迟、心房缺如、室间隔缺损、心室缺损等。结论血清碱性磷酸酶活性是反映体内锌水平的敏感指标,与血锌值结合能较准确地反映体内的锌水平。母体中等以上低锌能增加胚心畸形的发生,孕鼠低锌对胚胎的毒性作用存在较明显的剂量-效应关系。Objective To study developmental toxicity of zinc deficiency on embryonic heart of rat and investigate threshold dosage of toxic effect of zinc deficiency and dosage-effect relation. Methods 50 SD-healthy adult female rats (body weight :200-250g) were randomly divided into 5 groups: zinc severely deficient group and zinc deficient group; zinc moderately deficient group; zinc marginally deficient group and normal zinc level group and they were fed forages with different content of zinc. At first,they were fed for 25 days for depleting the zinc store in their body. Then they mated with male rats. All the pregnant rats were killed on the 19th day of pregnancy and heart malformation of embryos was observed through slices. Results As compared with normal zinc group, the serum AKP activity and serum zinc level of pregnant rats in all zinc deficient groups were significantly lower( for serum AKP activity: t = 86.79, 52.37, 38.32 and 13.35 respectively,all P 〈 0.05;for serum zinc level: t = 49.44, 27.92, 21. 10 and 8. 14 respectively, all P 〈 0.05 ). Under the same conditions, the changes in serum AKP activity were more significant. As compared with normal zinc group, the rates of embryo heart malformation in zinc severely deficient group, zinc deficient group and zinc moderately deficient group were significantly higher (X2 = 35.62, 16.93 and 5.91respectively, all P 〈 0.05 ), but there was no significant difference in incidence rate of embryo heart malformation between the zinc marginally deficient group and the normal zinc level group ( P 〉 0.05 ). Embryo heart malformations mainly included developmental delay of embryo heart, atrial absence, ventricular septal defect, ventricular defect and so on. Conclusion Serum AKP activity is a sensitive index of zinc level in body, if combined with the serum level of zinc it can more accurately reflect the level of zinc.
