摘  要:目的探讨神经调节蛋白-1β(neuregulin-1β,NRG-1β)预处理对大鼠视网膜缺血再灌注损伤后视网膜细胞凋亡的影响以及最佳给药时间窗。方法借鉴线栓法制备大鼠脑缺血再灌注模型的方法,制备大鼠视网膜缺血再灌注模型。将36只Wistar大鼠随机分为正常对照组(A),缺血再灌注损伤组(B),NRG-1β预处理48h、24h、12h、6h组(C、D、E、F)6组(每组6只),B、C、D、E、F各组在视网膜维持缺血状态1h再灌注24h后处死,立即摘除眼球制作石蜡切片,用TUNEL法检测视网膜细胞凋亡的变化。结果凋亡细胞主要出现在视网膜内核层及神经节细胞层,而外核层几乎无阳性表达。A、B、C、D、E、F各组凋亡细胞指数分别为0、28.58±2.53、15.56±1.19、4.35±1.72、8.15±1.96、11.46±1.65,与B组相比,NRG-1β预处理各组大鼠视网膜细胞凋亡数目显著减少(P<0.05),以D组凋亡细胞指数最低。结论NRG-1β预处理对视网膜缺血再灌注损伤具有明显保护作用,以NRG-1β预处理24h视网膜保护效果最好。Objective To investigate the effects of neuregulin-1β pretreatment on retinal cell apoptosis after ischemia-reperfusion injury and to explore the best drug delivery time. Methods The method of thread embolism was used to prepare the model of cerebral ischemia-reperfusion, and prepare the model of retinal ischemiareperfusion. Thirty-six Wistar rats were randomly divided into six groups, normal group ( A ), ischemia-reperfusion group ( B ), neuregulin-1β pretreatment groups ( C, D, E, F) of which rats received pretreatment of neuregulin-1β at 48 h,24 h,12 h,5 h before the 1 h of retinal ischemia and the followed 24 hreperfusion respectively,5 rats in each group. Retinal paraffin sections of rats were stained with TdT-mediated dUTP nick end labeling (TUNEL)to examine apoptosis of retinal cells. Results Apoptotic cells mainly appeared in the inner nuclear layer and the ganglion cell layer, and hardly found in the outer nuclear layer. The apoptotic index in each group were 0,28.58 ± 2.53,15.55 ± 1.19, 4.35± 1.72,8.15 ±1.95, 11. 45 ± 1.65 ,respectively,and the numbers of apoptotic cells in each neuregulin-1β pretreatment group were significantly lower than that of group B (P 〈 0.05 ), and the apoptosis index of the group D was minimum. Conclusion Neuregulin-1β pretreatment can significantly protect the retina after ischemia-reperfusion injury, and the best drug delivery time is about 24 h before the ischemia.
