摘  要:目的:观察大鼠肝纤维化过程中结缔组织生长因子(CTGF)的表达,并探讨其与肠源性内毒素血症的关系。方法:以四氯化碳制作大鼠肝纤维化模型,设立正常对照组、肝纤维化2周组、4周组、6周组、8周组;模型形成时经大鼠腹主动脉采血,产色基质偶氮法鲎试剂定量测定血浆内毒素,取肝左叶行天狼猩红胶原染色评价肝纤维化程度,Western-blot检测CTGF在肝脏的表达。结果:实验性大鼠肝纤维化过程中第2周血浆内毒素含量即明显升高(P<0.05),肝组织CTGF的含量第4周明显升高(P<0.05),之后两者均渐进性升高且呈显著正相关(r=0.918,P<0.01)。结论:在四氯化碳制作的实验性肝纤维化过程中,肠源性内毒素血症可能是导致肝脏CTGF高表达进而促进肝纤维化的重要因素。Objective:To investigate the expression of connective tissue growth factor (CTGF)in rat liver fibrogenesis and its association with intestinal endotoxemia. Methods: The fibrotic model of rats was induced by subcutaneous injection of carbon tetrachloride. Thirty-seven male Wistar rats(200 g_+20 g) were randomly divided into control group(n=5), fibrotic 2 w, 4 w, 6 w and 8 w groups(n=8 per group). At the end of the experiment, rats were killed for collection of blood and liver tissue. Liver fibrosis was scored by image analysis of sirius red stain. The level of endotoxin was measured using a Limulus Amebocyte Lysate test kit. The intensity of the expression of CTGF protein in liver was assessed by Western-blot. Results: During the fibrogenesis, the levels of plasma endotoxin were markedly higher from the second week, while enhanced expression of CTGF protein in liver from the fourth week. After then both of them increased gradually and were significantly, positively correlated. Conclusion: In rat experimental hepatic fibrogenesis induced by carbon tetrachloride, intestinal endotoxemia maybe one of the factors leading to CTGF overexpression and promote hepatic fibrosis.
