摘  要:目的研究犬肺癌模型抑癌基因表达与支气管动脉造影表现的相关性,探讨肺癌早期诊断的新方法、新标准。材料与方法22只犬,经气管插管由同轴微导管定位向右肺膈叶灌注甲基胆蒽、二乙基亚硝胺和超液化碘油混悬液,诱导犬肺癌模型,在不同时期进行基因检测和影像、病理学检查。结果p53基因蛋白随诱导时间延长,阳性表达率逐渐升高,p16逐渐减低。CD31血管内皮标记表达阳性与支气管动脉造影所见肿瘤样血管及染色吻合。结论CD31血管内皮细胞标记阳性表达和支气管动脉造影发现的肿瘤血管及染色是肺癌早期诊断的参考指标;早期检测p53基因突变与p16基因失活对提高早期诊断率和5年治愈率有重要意义。Objective To discuss the relationship between suppression gene expression and bronchial arteriography on the model of canine lung cancer and explore the new diagnostic methods of early-stage lung cancer. Materials and Methods 22 dogs were respectively administrated suspension of lipiodol-ultrafluid mixed with 3-methylcholanthrene (MCA) and diethylnitrosamine ( DEN ). The suspension was injected into diaphragmic lobe of right lung with co-axial catheter through endotracheal intubation. Histopathologic and imaging examination were performed and p53, p16 gene expression were detected at different period. Results When induced carcinogenesis time was prolonged, the positive rate of p53 gene expression was gradually increased and p16 gene was gradually reduced. The expression of vascular endothelial marker CD31 was positive. The tumor vessels and staining were observed by bronchial arteriography. Conclusion The positive expression of CD31 vascular endothelial marker and tumor vessels and staining observed by bronchial arteriography are diagnostic indicators of early-stage lung cancer. The abnormal expression of P53 and P16 gene are significant for improving diagnostic rate of early-stage lung cancer.
