摘 要:目的:探讨CXC族趋化因子受体4(CXC chemokine receptor 4,CXCR4)小干扰RNA对人大肠癌细胞株SW480迁移能力的抑制作用。方法:将与CXCR4 mRNA互补的小干扰RNA(siRNA)及阴性对照Non-silencing dsRNA以阳离子脂质体包埋后转染人大肠癌细胞株SW480,采用逆转录-聚合酶链反应(RT-PCR)检测转染后SW480细胞CXCR4 mRNA表达的变化,采用Western blotting检测转染细胞CXCR4蛋白表达的变化,采用Transwell迁移实验检测大肠癌细胞SW480迁移能力的变化。结果:与正常对照组和Non-silencing dsRNA相比,siRNA各浓度组对SW480细胞CXCR4 mRNA和蛋白的表达均有明显的抑制作用(P<0.01),其抑制效应具有浓度依赖性(P<0.01)。与正常对照组和Non-silencing dsRNA相比,siRNA各剂量组对肿瘤细胞迁移有明显抑制作用(P<0.01),其抑制效应具有剂量依赖性(P<0.01);siRNA在终浓度为50、100和200 nmol/L时对SW480细胞的迁移抑制率分别为20.5%、37.5%和52.9%。结论:CXCR4小干扰RNA通过下调CXCR4 mRNA和蛋白的表达显著抑制人大肠癌细胞SW480的迁移,并呈剂量依赖性。Objective:To evaluate the inhibitory effect of CXCR4-targeted small interfering RNA (siRNA) on migration of human coloreetal cancer cell line SW480. Methods: The siRNA targeting CXCR4 mRNA and non-silencing double-stranded RNA (Non-silencing dsRNA) were transfected into SW480 cells via liposome Lipofectin. The total RNAs and proteins were extracted from the cells 48 h after transfection; semi-quantitative RT-PCR and Western blotting were performed to evaluate the levels of CXCR4 gene and protein expression, respectively. The migration ability of transfected SW480 cells were measured quantitatively by Transwell migration assays. Results: The expression of CXCR4 gene and protein and migration of SW480 cells treated with different final concentrations of siRNA were significantly decreased compared with those of the normal control and the non-silencing dsRNA group (P 〈 0.01 ). Besides, the inhibitory effects were significantly different between the cells treated with different concentrations of siRNA (P 〈 0.01 ). The migration inhibition rates were 20.5% , 37.5% and 52.9% when the final concentrations of siRNA were 50, 100 nmol/L and 200 nmol/L, respectively. Conclusion: CXCR4 siRNA targeting CXCR4 mRNA can significantly inhibit the migration of human coloreetal cancer cell line SW480, and the inhibition is in a dose-dependent manner.
