子宫肌瘤组织17β-羟类固醇脱氢酶活性的测定

摘  要:为研究17β-羟类固醇脱氢酶(17β-HSD)与子宫肌瘤的关系,用3H-E2(终浓度0.5mol/L)作底物,以3H-E2氧化为3H-E1的量表示17β-HSD的活性,经薄层层析后分别测定3H-E2、3H-E1的每分记数。结果显示,子宫内膜增殖期肌瘤组织E2含量低于正常肌层,而分泌期高于子宫肌层,但差别无显著性;分泌期子宫肌瘤E2转换率最低,提示此时17β-HSD活性最低。33.33%(4/12)肌瘤患者子宫内膜反应不符合正常月经周期表现,41.6%(5/12)患者伴卵巢滤泡囊肿,91.6%(11/12)属多发性子宫肌瘤。表明E2含量与17β-HSD活性改变有关,是子宫肌瘤发生。Leiomyoma was surgically removed from 12 patients aging from 31 to 60 pathological examination.The tissue was homogenized and centrifuged at 800×g for 10 mins (performed at 0 ̄4℃).The supernatant 200μl were incubated with NAD +(0.5mmol/L)buffer 200μl, 3H E 2(final concentration 0.5mol/L)20μl,at 37℃ for 30 min.The reaction was stopped by the addition of a 5 fold volume of ether. After extraction,the steriods containing unlabled E 1 and E 2 carriers were isolated by thinlayer chromatography in a choloroform cyclohexane system (V/V 8:2).The amount of E 1 formed from E 2 was calculated by dividing the amount of 3 H Ε 1 by the specific activity of 3 H E 2 as substrate. During menstrual cycle the 17β HSD assay showed the same enzyme activity at proliferative phase in normal and leiomyoma tissues,but lower activity in two tissues at secretory phase,especially in leiomyoma tissues.The conversion rate of E 2 to E 1 was expressed as picomoles of E 1 formed per μg protein.The conversion rate in secretory phase was the lowest in leiomyoma tissues (14.82pg/μg protein).91.60%(11/12)patients had multiple leiomyoma and 41.60%(5/12)had ovary nabothian cyst.Some patients' endometrium showed secretory phase in proliferative phase of menstruation or otherwise.These results showed that the activition of 17β HSD is one of the reasons in increasing the level of E 2 at secretory phase.
