摘 要:为探讨孤儿核受体(Nurr1)在多巴胺能神经元损伤修复过程中对特异性表型酪氨酸羟化酶(TH)的影响,本研究用6-羟基多巴胺(6-OHDA)损伤大鼠一侧前脑内侧束(MFB)制备单侧帕金森病(PD)模型,分别应用Western blot和免疫组织化学技术观察不同时间点中脑黑质中Nurr1、TH蛋白表达的变化及其相关性。结果显示,Nurr1蛋白表达在术后1d损伤侧黑质中即明显下降,7d后开始逐渐上升,14d接近正常水平,28d明显升高;TH蛋白表达在术后1d损伤侧黑质中无明显变化,7d开始下降,14d达最低,28d开始上升。上述结果提示,在6-OHDA损伤黑质-纹状体通路后,Nurr1的表达先下降后上升,其表达变化早于TH,可能在PD模型大鼠的损伤修复中发挥重要作用。To study the effect of Nurr1 on the specific phenotype TH in the injurious and restorative process of dopaminergic neurons, 6- OHDA was injected into the light MFB of adult rats to construct unilateral PD model, and Western blot and immunohistochemistry were used to observe the expressive changes of Nun'l and TH protein and their relativity in the substantia nigra at different times. The results showed that the Nurr1 protein expression was apparently decreased in the leisoned substantia nigra on the 1 st day after operation, started to increase gradually on the 7th day, approached the normal level on the 14th day, and markedly increased on the 28th day. However, the TH protein expression was of little change in the leisoned substantia nigra on the 1 st day after operation, started to decrease on the 7th day, reached the bottom on the 14th day, and started to increase on the 28th day. The results above suggest that after the injury of substantia nigra-striatum pathway, the Nurr1 expression decreases earlier and increases later, which occurrs prior to TH alteration, suggesting that this protein may play an important role in the injurious and restorative process of PD model rats.
