摘 要:用放射免疫法和高效液相色谱电化学检测器分别测定了SAMP8小鼠血浆、大脑皮层、海马、下丘脑皮质酮(corticosterone,CORT)水平和大脑皮层、海马兴奋性氨基酸含量。结果显示,SAMP8小鼠血浆CORT水平均显著(P<0.05,或P<0.01)高于其同龄正常对照小鼠SAMR1。SAMP8小鼠8月和12月龄大脑皮层和海马CORT水平也明显(P<0.05,或P<0.01)高于同龄正常对照SAMR1小鼠。同时,SAMP8小鼠大脑皮层和海马谷氨酸(glu-tamate,Glu)含量有上升趋势,谷氨酰胺(Glutamine,Gln)含量则明显高于SAMR1小鼠。SAMP8小鼠切除肾上腺后,大脑皮层和海马Glu含量呈下降趋势,而Gln含量则显著降低。给切除肾上腺的SAMP8小鼠补充CORT(ip,10mg/kg),大脑皮层和海马Glu和Gln水平均明显(P<0.05,或P<0.01)升高,而天冬氨酸(Aspartate,Asp)含量始终没有变化。结果提示,SAMP8小鼠大脑皮层和海马Glu和Gln水平升高是肾上腺依赖的。SAMP8小鼠大脑皮层和海马Glu和Gln含量升高可能与其大脑皮层和海马CORT水平升高有?Plasma corticosterone (CORT) concentration in SAMP8 increased significantly ( P <0. 05, or P <0. 01 ) compared with age matched SAMR1 while the contents of CORT in both cerebral cortex and hippocampus were also markedly ( P <0. 05 ,or P <0. 01) greater in 8 month and 12 month old SAMP8 than those in age matched SAMR1. Meanwhile, contents of glutamate (Glu) in cerebral cortex and hippocampus had a trend to rise and contents of glutamine (Gln) increased pronouncedly in SAMP8 compared with those in SAMR1. However, adrenalectomy in SAMP8 led to significant reduction of Gln contents in cerebral cortex and hippocampus while Glu contents decreased without statistical significance. Supplementation with CORT to adrenalectomized SAMP8 could once again result in a great elevation in both Glu and Gln contents. The present results suggest that the elevated contents of Glu and Gln in cerebral cortex and hippocampus in SAMP8 are adrenal dependent and the elevation of CORT levels in both cerebral and hippocampus in SAMP8 may be responsible for the elevated contents of Glu and Gln.
