摘 要:目的研究酸性肽对AD大鼠脑中NGF、NMDAR1表达的影响。方法采用大鼠脑组织立体定位微量注射技术,建立AD动物摸型,用不同剂量的AP对AD大鼠进行治疗。通过测定与学习记忆有关的行为学指标及脑内与AD相关的NGF、Aβ水平的变化,并与正常组、模型组、生理盐水组结果比较,对数据进行统计处理。结果模型组与正常组比较,学习记忆成绩明显下降(P<0.01);各治疗组与模型组比较,学习记忆成绩明显提高(P<0.01);模型组与空白组比较,大鼠基底前脑中NGF水平显著降低,皮质中NMDAR1含量显著增加,各治疗组分别与模型组比较,大鼠基底前脑中NGF水平显著降低,皮质中NMDAR1含量显著增加(P<0.01)。结论AP可能通过上调AD基底前脑中NGF的水平并降低NMDAR1水平来发挥对AD的治疗作用。Objective To investigate the effect of acidic neuropeptide on learning and remembering functions and levels of NGF, NMDAR1 in brain of AD rat . Methods The AD animal model was established by injecting IBO 1ul (5ug) into rats brain to damage bilateral nbM in rat brain. The AD rats were treated with AP. The experiment lasted for 20 days , the drugs were given once a day. The learning and remembering ability was tested by Y- maze test. The levels of nerver growth factor (NGF) in base of forebrain and NMDAR1 in cerebral cortex were determined according to the procedures of immunohistochemistry. Results Compared with normal group,the learning and remembering ability of model groups decreased significantly(P〈 0.01), Comparing with model group, the learning and remembering ability of AP groups increased significantly(P〈0.01) . NGF levels in AP-30 mg/(kg·d) group and AP-60 mg/(kg· d) group increased significantly(P〈0.01). The levels of NMDAR1 in AP-30 mg/(kg·d) group and AP-60 mg/(kg·d) group decreased significantly(P〈0. 01). Conclusion AP can cure AD by up-regulating the level of NGF in base of forebrain and down-regulating the levels of NMDAR1 and Aβ in cerebral cortex.
