血管内注射β淀粉样蛋白1-40后大鼠脑实质超微结构及空间学习和记忆能力的改变

摘　　要：目的研究血管内注射可溶性β淀粉样蛋白_(1-40)(Aβ_(1-40))后,对大鼠脑实质超微结构的破坏作用及空间学习和记乙能力的影响。方法 SD大鼠78只,随机分为Aβ_(1-40)组和对照组,每组39只,每组再根据注射后饲养时间的不同分为15、20、30天3个时间点,各组行Morris水迷宫进行空间学习和记忆能力的测试,然后麻醉灌注,处死大鼠取脑,并行电镜观察。结果 Aβ_(1-40)组和对照组在15、20、30天不同训练时间段分别比较,Morris水迷宫各项行为学指标改变不显著。电镜下观察,Aβ_(1-40)组血脑屏障内血管内皮细胞、星形胶质细胞的脚板,脑实质内神经元、星形胶质细胞、小胶质细胞出现不同程度的改变。结论经过14天的注射,血液中Aβ_(1-40)对大鼠脑实质有着广泛的毒性损害作用,但是对行为学改变的影响不显著。Objective To study damages of rats brain ultrastructure and spatial learning and memory ability after intravascutar injection of soluble Aβ1-40. Methods Seventy-eight male SD rats were divided into 2 groupsAβ1-40 group and normal saline group） randomly,each group was divided again into 3 subgroups according to the time after injection. The animals were anesthetized with sodium pentobarbital so that a polyethylene cannula could be inserted into the right external jugu- lar vein to approaching or entering the right atrium. On the day following eannulation,the Aβ1-40 groups began receiving twice daily intravenous injection of human Aβ1-40 The normal saline groups served as controls and received twice daily intravenous injection of normal saline. Then morris water maze was used to test the cognition of the rats, after which, the rats were sacrificed and the brains were examined by electron microscope. Results In morris water maze test, there was no significant difference in overall acquisition between the two groups （P〉0.05）. However, electron microscope examination showed that Aβ1-40 caused changes in the endothelial cells, the end feet of the astroeytes and basement membrane of the BBB,and also could do damage to many cells in the brain,such as the microglia,astrocytes and the neurons. Conclusion Intravascular injection of Aβ1-40 will do damage to the brain of rats.
