应用组织芯片技术研究间变性大细胞淋巴瘤中ALK蛋白的表达和意义

摘　　要：目的:应用组织芯片技术研究间变性大细胞淋巴瘤(ALCL)中ALK蛋白的表达及其与患者年龄和预后的关系。方法:建立包含30例ALCL及2例对照组织共96点阵的组织芯片,采用免疫组织化学SP法检测ALK蛋白表达,并应用SPSS软件进行统计学分析。结果:(1)ALK蛋自在2例皮肤原发ALCL均为阴性,28例系统性ALCL中有20例(71.4%)阳性,主要为细胞浆和/或细胞核棕黄色。(2)ALK蛋白阳性组的年龄低于ALK蛋白阴性组的年龄,两者差异具有显著性(P<0.05)。(3)ALK蛋白阳性组的预后好于ALK蛋白阴性组的预后,两者差异具有显著性(P<0.05)。结论:ALK蛋白的表达在ALCL中有很高的发生率,尤其是低年龄组的患者,它可以成为ALCL的诊断、鉴别诊断和判断预后的一个独立重要指标。ObJective： To investigate the relation of the anaplestic lymphoma kinase （ALK） protein、 age and prognostic in anaplastic large cell lymphoma by tissue microarray technique. Method： 30 cases of anaplastic large cell lymphoma and 2 cases d normal control tissues were examined using tissue microarray technique by immunohistochemistty, together with analysis by statistics. Resalts： （1） ALK protein expressed negative in 2 samples of primarily ALCL, but 9 out of 19 systematic ALCL expressed ALK protein in cytoplasm and/or nucleus （71.4%） . （2） CLinicaLly, the patients with ALK expression were younger than those without ALK expression （P〈0.05） . （3） The prognosis with ALK expression was better than those without p80 expression （P 〈0.05） . Conclusion： There was a high incidence rate of ALK expression in ALCL, especially in younger group. ALK expression might be an useful and independent marker for the differential diagnosis and prognosis estimation of ALCL.
