摘  要:目的:探讨碱性成纤维细胞生长因子(basic fibroblast growth factor,bFGF)反义寡核苷酸对肾癌细胞凋亡的影响,以期寻找肾癌反义治疗的合适药物。方法:以脂质体作为转染载体,将bFGF反义寡核苷酸转染人肾癌细胞系786-0,应用RT-PCR和Western印迹法检测转染bFGF反义寡核苷酸后肾癌细胞中bFGF mRNA及其蛋白的表达,应用流式细胞仪检测转染bFGF反义寡核苷酸后肾癌细胞的凋亡率。结果:与转染错义寡核苷酸组相比,转染bFGF反义寡核苷酸组786-0细胞中bFGF mRNA及其蛋白的表达有明显下降,且转染bFGF反义寡核苷酸可明显提高786-0细胞的凋亡率(P<0.01)。结论:经bFGF反义寡核苷酸作用后,肾癌细胞bFGF的表达受到了抑制,同时促进了肾癌细胞的凋亡,因此应用bFGF反义寡核苷酸治疗肾癌可能是一种较有希望的肿瘤治疗新方法。Objective: To investigate the effects of antisense oligodeoxynucleotide targeting basic fibroblast growth factor (bFGF) on apoptosis of human renal carcinoma cell line and find the appropriate agents for renal carcinoma therapy. Methods: The antisense bFGF oligodeoxy-nucleotides were transfeeted into human renal carcinoma cell line 786-0 by lipofeetin. Inhibition of bFGF mRNA and protein expressions was investigated by RT-PCR and Western blotting after transfection, respectively. The apoptosis of transfected cells was determined by flow cytometric analysis. Results: The bFGF mRNA and protein expressions were significantly decreased after transfection with antisense bFGF oligodeoxynucleotides compared with those transfeeted with missense bFGF oligodeoxy- nucleotides. Transfection with antisense bFGF oligodeoxynucleotides markedly increased the apoptotic rate of 786-0 cells ( P 〈0. 01 ). Conclusion: Treatment with antisense bFGF oligodeoxynucleotides inhibited the expressions of bFGF at mRNA and protein levels and increased the apoptosis of renal carcinoma 786.0 cells, Antisense bFGF oligodeoxynucleotides may be a desirable novel approach for treatment of renal carcinoma.
