摘 要:目的:观察血管内皮生长因子(vascular endothelial growth factor,VEGF)反义寡核苷酸对白血病细胞系K562细胞凋亡的作用。方法:实验分为VEGF反义组、VEGF错义组和空白对照组,应用阳离子聚合物介导硫代修饰VEGF寡核苷酸转染体外培养的白血病细胞系K562细胞48h后,观察细胞形态,琼脂糖凝胶电泳法检测DNA梯状带,流式细胞仪AnnexinⅤ-FITC/PI检测细胞凋亡。结果:反义组可见细胞集落减少,细胞皱缩,细胞质中颗粒增多,核固缩并出现细胞碎片;而错义组和空白对照组细胞均悬浮成团,细胞轮廓清楚,胞体透亮,生长旺盛。反义组见凋亡梯形带,错义组和空白对照组仅1条DNA条带。反义组细胞凋亡率达23.28%,与错义组和空白对照组相比差异具有统计学意义(P<0.05)。结论:VEGF反义寡核苷酸可诱导白血病细胞凋亡,提示VEGF与抗白血病细胞凋亡有关。Object:To observe the effect of antisense oligodeoxynucleotides (ASODN) of vascular endothelial growth factor (VEGF) on apoptosis of human leukemic cell line K562 ceils. Methods: The cells were divided into VEGF ASODN group, VEGF missense ODN (MSODN) group, and empty control group. K562 cells were treated with phosphorothioate-modified antisense VEGF ODN formulated in cationic polymer in vitro for 48 h. The cell morphology was observed. DNA ladder was detected by gel electrephoresis. Apoptosis was detected by flow cytometric analysis ( Annexin V-FITC/PI assay). Results : In the ASODN group, the number of cell clusters was decreased and typical apoptosis morphological changes appeared, such as shrinkage of cell bodies, more granular structure in cytoplasm, pyknosis, and DNA fragmentation. In VEGF MSODN group and non transfection group, cells were clustering in suspen- sions with distinct boundaries and transparent bodies and proliferated prominantly. The DNA ladder was observed in the VEGF ASODN group. But there was only one strand of DNA in the MSODN group and the control group. The apoptotic rate in the ASODN group was 23.28%, which was significantly higher than that in the MSODN group and the control group ( P 〈 0.05 ). Conclusion: VEGF ASODN could induce apoptosis of leukemia cells. VEGF was related with anti-apoptosis effect on leukemia cells.
