PGI_2/TxA_2、tPA/PAI动态平衡与黑色素瘤B_(16)侵袭率的关系

摘　　要：目的了解肿瘤侵袭率与ＰＧＩ２／ＴｘＡ２和ｔＰＡ／ＰＡＩ平衡的关系。方法参照Ｎｉｃｏｌｓｏｎ报道的方法建立体外黑色素瘤Ｂ１６侵袭血管内皮细胞单层模型，利用放射免疫酶联技术检测肿瘤侵袭过程中，ＰＧＩ２、ＴｘＡ２、ｔＰＡ、ＰＡＩ时相性分泌变化，以探究ＰＧＩ２／ＴｘＡ２、ｔＰＡ／ＰＡＩ比值变化，在肿瘤侵袭和转移过程中的作用。结果在肿瘤侵袭过程中，总ＰＧＩ２、ＰＡＩ分泌量呈下降趋势，ＴｘＡ２和ＰＡＩ分泌呈上升趋势；黑色素瘤Ｂ１６细胞能够通过分泌ｔＰＡ、ＰＡＩ、ＰＧＩ２和ＴｘＡ２来破坏血液循环系统中ＰＧＩ２／ＴｘＡ２和ｔＰＡ／ＰＡＩ平衡；ＰＧＩ２／ＴｘＡ２、ｔＰＡ／ＰＡＩ比值变化，肿瘤侵袭率呈对应性变动。结论ＰＧＩ２／ＴｘＡ２和ｔＰＡ／ＰＡＩ分泌的平衡失调可能是肿瘤侵袭和转移形成的重要原因之一。URPOSE To study the effect of PGI2/TxA2 and tPA/PAI balance on tumor invasion.METHODS Tumor cell invasion model was established in vitro, and the ratio of phasic invasion was measured meanwhile radioimmunology and enzyme labelling techniques were used to record changes of phasic secreton of PGI2, TxA2, tPA and PAI before and after endothelial cells were exposed to 15 Gy γradiation.RESULTS ①The ration of TxA2/PGI2, tPA/PAI secreted by B16 cell was larger than that of the endothelial cells; ②In the invasion process PGI2/TxA2 decreased, while tPA/PAI increased; ③After endothelial cells were exposed to 15 Gy γradiation, tPA/PAI decreased and PGI2/TxA2 decreased; ④In the control invasion process and during the time after endothelial cells were exposed to radiation, the phasic change of invasion ratio corresponds with the phasic change of total secretion of PGI2, TxA2, tPA and PAI.CONCLUSIONS The effect of PGI2/TxA2 and tPA/PAI balance play an important role on the tumor invasion.
