摘 要:目的了解肿瘤侵袭率与PGI2/TxA2和tPA/PAI平衡的关系。方法参照Nicolson报道的方法建立体外黑色素瘤B16侵袭血管内皮细胞单层模型,利用放射免疫酶联技术检测肿瘤侵袭过程中,PGI2、TxA2、tPA、PAI时相性分泌变化,以探究PGI2/TxA2、tPA/PAI比值变化,在肿瘤侵袭和转移过程中的作用。结果在肿瘤侵袭过程中,总PGI2、PAI分泌量呈下降趋势,TxA2和PAI分泌呈上升趋势;黑色素瘤B16细胞能够通过分泌tPA、PAI、PGI2和TxA2来破坏血液循环系统中PGI2/TxA2和tPA/PAI平衡;PGI2/TxA2、tPA/PAI比值变化,肿瘤侵袭率呈对应性变动。结论PGI2/TxA2和tPA/PAI分泌的平衡失调可能是肿瘤侵袭和转移形成的重要原因之一。URPOSE To study the effect of PGI2/TxA2 and tPA/PAI balance on tumor invasion.METHODS Tumor cell invasion model was established in vitro, and the ratio of phasic invasion was measured meanwhile radioimmunology and enzyme labelling techniques were used to record changes of phasic secreton of PGI2, TxA2, tPA and PAI before and after endothelial cells were exposed to 15 Gy γradiation.RESULTS ①The ration of TxA2/PGI2, tPA/PAI secreted by B16 cell was larger than that of the endothelial cells; ②In the invasion process PGI2/TxA2 decreased, while tPA/PAI increased; ③After endothelial cells were exposed to 15 Gy γradiation, tPA/PAI decreased and PGI2/TxA2 decreased; ④In the control invasion process and during the time after endothelial cells were exposed to radiation, the phasic change of invasion ratio corresponds with the phasic change of total secretion of PGI2, TxA2, tPA and PAI.CONCLUSIONS The effect of PGI2/TxA2 and tPA/PAI balance play an important role on the tumor invasion.
