摘 要:目的探讨乐尔脉对脑缺血再灌注损伤中期大鼠海马神经细胞凋亡和Fas、Bax表达的作用与机制。方法采用大鼠右侧大脑中动脉内栓线阻断法(MCAO)造成局灶性脑缺血再灌注模型,凋亡细胞原位末端标记法(TUNEL)检测海马神经细胞凋亡,免疫组化与逆转录聚合酶链反应(RT-PCR)技术检测大鼠海马Fas、Bax、Caspase-3、Caspase-9mRNA的表达,并进行图像处理分析结果。结果在缺血再灌注损伤海马组织可检测到较多的凋亡细胞,主要分布在CA1区。海马FasmRNA表达上调,海马CA1区锥体层神经元、胶质细胞、室管膜细胞Fas蛋白表达均明显增加,与凋亡呈同样趋势。海马Bax mRNA上调,但与假手术组比较无统计学意义(P>0.05),Bax蛋白水平却显著增加,下游Caspase-3、Caspase-9mRNA显著上调。乐尔脉和氟桂利嗪可明显降低缺血再灌注损伤大鼠海马神经元细胞凋亡,降低Fas mRNA和Fas蛋白表达,但不降低Bax mRNA和Bax蛋白表达。结论乐尔脉限制和减少脑缺血再灌注损伤中期神经细胞凋亡的发生和发展,对局灶性脑缺血再灌注损伤具有保护作用,其作用机制可能与乐尔脉降低海马Fas mRNA表达而抑制神经细胞凋亡有关。Objective To study the effects of Le'ermai on apoptosis of hippocampal neuronal cell in cerebral ischemic reperfussion injured rats. Methods Experimental models of focal ischemia reperfussion injury rats were established by intraluminal occlusion of the middle cerebral artery(MCAC) in rat. TUNE1. staining methods were used to detect the characteristic of the apoptotic cell death. Immunohistochemical and RT- PCR technique were employed to detect the transcription of Fas mRNA, Bax mRNA in hippocampus during reperfusion for 10 day after 2 h ischemia. Results The apoptotic ceils were examined in hippocampus tissue and dissemination in hippocam ous CA1 area. The transcription of FasmRNA was up - regulated, the expression of Fas protein in pyramidel laminal neuron, ependymal cell and neuroglia were significantly increased. Up -regulation of transcription of caspase- 3, 9 mRNA in hippocampus of cerebral iscbemic reperfusion injury was observed. Le'ermai could significantly decrease the apoptosis of hippocampus neurons and the expression of fas , whereas inhibitory effect on Bax mRNA was not examined. Conclusion Le'ermai limited or decreased deve lopment of hippocampus neuronal apoptosis in medium period of brain ischemia reperfusion, which relieved brain damage induced by focal brain ischemia reperfusion. The mechanism of protective effect of Le'ermai might be related to lowering transcription of fas mRNA and inhibiting apoptosis of hippocampus.
