室管膜前下区神经干细胞诱导分化为γ-氨基丁酸能神经元的实验研究

摘  要:目的研究骨形成蛋白-2(bone morphogenetic protein-2,BMP2)及星形胶质细胞条件培养液(astrocyte-conditioned medium,ACM)在室管膜前下区(anterior subventricular zone,SVZa)神经干细胞分化为γ-氨基丁酸(gamma-aminobutyric acid,GABA)能神经元中的作用。方法体外培养SVZa神经干细胞,分为对照组、BMP2组、ACM组、BMP2+ACM组分别予以诱导分化,通过免疫组化和流式细胞技术分别检测各组SVZa神经干细胞分化后GAD67+细胞的比例。结果各实验组的GAD67+细胞的比例均明显高于对照组,其中BMP2+ACM组尤其显著,差异有显著性(P<0.05)。结论BMP2及ACM可以促进SVZa神经干细胞向GABA能神经元分化,而且BMP2与ACM有正协同作用。[Objective] To study the role of bone morphogenetic protein-2 (BMP2) and astrocyte-conditioned medium (ACM) in SVZa neural stem cells differentiating into GABAergic neurons. [Methods] The neural stem cells of SVZa were separated and cultured in vitro, which were divided into: Control group, BMP2 group, ACM group, BMP2+ACM group. The GAD67+ ceils rate of differentiation of the neural stem ceils of SVZa was detected by immunohistochemistry and flow cytometry (FCM). [Results] The rate of GAD67+ cells differentiation in each experiment group were much higher than Control group, especially in the group of BMP2+ACM. There was significant difference between all groups by analysis of variance (P 〈0.05). [Conclusion] BMP2 and ACM can promote the GABAergic neuronal differentiation of the neural stem ceils of SVZa and they were positive cooperativity.
